摘要
目的研究海莲叶乙醇提取物(BLE60)的抗氧化和抑制黑素作用,并对它的皮肤刺激性进行评价。方法首先通过检测二苯代苦味酰肼(DPPH)自由基和细胞活性氧(ROS)研究BLE60的抗氧化活性,接着在小鼠B16黑色素瘤细胞上研究BLE60对酪氨酸酶和黑素的抑制作用,然后以Western blotting检测BLE60对酪氨酸酶表达的影响,最后通过家兔皮肤多次给药研究BLE60的皮肤刺激性。结果 BLE60能清除DPPH自由基和UVA诱导的ROS,同时也抑制细胞酪氨酸酶活性、减少黑素产生和酪氨酸酶表达。BLE60多次给药对家兔皮肤无刺激性。结论 BLE60具有抗氧化和抑制黑素活性,且对皮肤无刺激性。BLE60主要通过下调酪氨酸酶表达来降低酪氨酸酶活性,从而抑制黑素产生。
        Objective To study the antioxidative and melanin-inhibitory effects of alcohol extract of Bruguiera sexangulaleaves(BLE60),and to evaluate its dermal irritation.Methods Firstly,antioxidative effects of BLE60 were explored by measuring 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical and intracellular reactive oxidative species(ROS).Secondly,inhibitory effects of BLE60 on tyrosinase and melanin were investigated in mice B16 melanoma cells.And then Western blotting was performed to detect tyrosinase expression.Finally BLE60 was repeatedly administered on rabbit skin to evaluate its dermal irritation.Results BLE60 inhibits the activity and expression of tyrosinase,and reduces melanin production as well.The extract which scavenges DPPH radical and ROS does not show dermal irritation in repeated administration.Conclusion BLE60 has activities of antioxidation and inhibiting melanin production,whereas has no dermal irritation.BLE60 inhibits melanin production mainly by decreasing tyrosinase activity,which results from down-regulation of the enzyme expression.
