摘要
目的探讨长链非编码RNA(lncRNA)肺腺癌转移相关转录本1(MALAT1)与乳腺癌淋巴结转移的关系。方法收集新疆医科大学附属肿瘤医院乳腺外科一病区2018年10-12月收治的乳腺癌住院患者20例,采用乳腺手术+前哨淋巴结活检术,病理诊断为乳腺浸润性癌,用前哨淋巴结(SLN)探测仪,术中冰冻示SLN有转移条件的患者对腋窝淋巴结(ALN)进行常规的清扫。获得同时满足SLN(+)及ALN(+)的乳腺癌患者12例,分别取SLN、ALN及乳腺癌组织、癌旁组织。采用Trizol法提取各组织的总RNA,采用分光光度仪定量计算RNA,进行RNA反转录,采用RT-PCR法检测lncRNA的表达,用2~(-△△ct)法计算LncRNA MALAT1的相对表达量。结果各组织中MALAT1相对表达量有明显差异(P<0.05),MALAT1在SLN中的表达明显高于癌旁组织与癌组织(P<0.05)。乳腺癌组织中MALAT1的表达量明显高于癌旁组织和ALN组织(P<0.05)。MALAT1表达与淋巴结转移率呈正相关(r=0.784,P<0.05),但与远处转移和淋巴结转数目不相关。结论 MALAT1在乳腺前哨淋巴结及癌组织中有异常高表达,其检测对乳腺癌淋巴结转移有一定的价值。
        Objective To investigate the relationship between lcnRNA of lung adenocarcinoma metastasis-associated transcript 1(MALAT1) and lymph node metastasis in breast cancer. Methods Twenty patients with breast cancer admitted to the Department of Breast Surgery, the Affiliated Tumor Hospital of Xinjiang Medical University from October to December 2018 were enrolled. All 20 patients underwent breast surgery and sentinel lymph node(SLN) biopsy. For breast invasive carcinoma, surgically removed primary tumor tissue and matched paracancerous tissue were treated with a sentinel lymph node detector. SLN≥2 was detected for methylene blue dye and frozen during surgery. Patients with metastatic SLN were treated with conventional axillary lymph node(ALN). 12 patients with breast cancer who met SLN(+) and ALN(+) were enrolled in. SLN, ALN, breast cancer tissues and paracancerous tissues were collected to be extracted the total RNA by Trizol method. The photometric apparatus performed quantitative calculation of RNA, performed reverse transcription of RNA, detected the expression of lncRNA by RT-PCR, and calculated the relative expression of lncRNA of MALAT1 by 2~(-ΔΔct) method. Results The relative expression of MALAT1 in every tissue was statistically significant(P<0.05). The expression of MALAT1 in SLN was significantly higher than that in adjacent tissues and cancer tissues(P<0.05). MALAT1 in breast cancer tissues was significantly higher than that in adjacent tissues and ALN tissues(P<0.05). There was a positive correlation with lymph node metastasis rate, high lymph node metastasis rate, and higher expression of MALAT1(r=0.784, P<0.05), but not related to distant metastasis and lymph node number(P>0.05). Conclusion MALAT1 is highly expressed in the sentinel lymph nodes and cancer tissues of the breast. MALAT1 detection has certain value for lymph node metastasis of breast cancer.
