摘要
目的在现有测定方法上,改进保健食品中测定EPA、DHA和α-亚麻酸的前处理方法,并进行方法学研究。方法样品经皂化、甲酯化后加正己烷定容,加氯化钠溶液混匀后离心静置,取上清液测试,经DB-FFAP毛细管色谱柱分离测定。结果该方法在50-850μg·mL-1间具有良好的线性(r>0.999),检出限为0.8~1.5mg·kg-1,回收率90%~105%,相对标准偏差<5%。结论本方法可用于鱼油保健品中EPA、DHA和α-亚麻酸的同时测定,操作简单快捷,结果可靠,重现性高。
OBJECTIVE To optimize the pretreatment methods for simultaneous determination of EPA(Eicosapentaenoic acid),DHA(Docosahexaenoic acid) and α-Linolenic acid in fish oil by GC,and study on methodology.METHODS DHA,EPA and α-Linolenic acid were extracted by saponification and esterification,added hexane to the mark,then added sodium chloride solution,mixed,centrifuged and stand seconds,analyzed by gas chromatography coupled with DB-FFAP capillary column used to separate the fatty acids.RESULTS The mass concentration of DHA,EPA and α-Linolenic acid were linear with peak area in the range of 50~850μg·mL-1,and regression coeficient was not less than 0.999.The average rate ranged from 90% to 105%,and the relative standard deviation was not more than 5%(n=3).CONCLUSION The method is simple and convenient,it can be used to determine the content of DHA,EPA and α-Linolenic acid in fish oils with good accuracy and repeatability.
