摘要
研究液体发酵桑黄菌落所提取的黄酮抗肿瘤活性。通过乙酸乙酯萃取液体发酵桑黄菌,纯化后得到总黄酮。采用MTS法检测总黄酮的细胞活力及对小鼠成纤维细胞L929细胞及肿瘤细胞HepG-2细胞的抑制情况,并使用流式细胞仪测定不同质量浓度总黄酮(10、20、50、100μg/m L)对正常细胞及肿瘤细胞的周期影响。结果发现,药用真菌桑黄菌液体发酵所提取的不同质量浓度总黄酮(10、20、50、100μg/m L)对小鼠成纤维细胞L929细胞无抑制作用,对HepG-2细胞有直接抑制作用。液体发酵的桑黄菌落所提取的总黄酮对小鼠肿瘤有抑制作用,并且具有剂量依赖性。
        To study the anti-tumor activity of flavonoids extracted from liquid fermented Phellinus igniarus,the liquid flavonoids were extracted by ethyl acetate and purified to obtain total flavonoids. MTS method was used to detect the cell viability of total flavonoids and the inhibition of mouse fibroblasts L929 cells and tumor cells HepG-2 cells. Flow cytometry was used to determine the effects of the total flavonoids in different concentrations( 10,20,50,100 μg/m L) on the cycle of normal cells and tumor cells. The results show that the different concentrations of total flavonoids( 10,20,50,100 μg/m L) extracted from liquid fermentation of Phellinus igniqrus had no inhibitory effect on mouse fibroblast L929 cells and had a direct inhibitory effect on HepG-2 cells. The total flavonoids extracted from liquid fermented Phellinus igniarius can inhibit the tumorigenesis of mice in a dose-dependent manner.
