健脾化痰方对裸鼠前胃癌MFC瘤组织P53 Bcl-2 Survivin Bax基因表达的影响

摘要
目的观察健脾化痰方对前胃癌(MFC)裸鼠肝转移模型瘤组织中P53,Bcl-2,Survivin,Bax基因表达的影响,探讨健脾化痰方抗肿瘤作用的分子机制。方法将MFC细胞接种于裸鼠脾脏中,建立裸鼠肝转移模型后,随机分为模型组、氟尿嘧啶(5-Fu)组及健脾化痰方高、中、低剂量组,给药4周后处理动物,用免疫组织化学法、RT-PCR法分别检测P53,Bcl-2,Survivin,Bax蛋白及mRNA表达。结果健脾化痰方各组均能下调P53,Bcl-2,Survivin蛋白及mRNA的表达,上调Bax蛋白及mRNA的表达。结论健脾化痰方的抗胃癌作用与下调P53,Bcl-2,Survivin的表达,上调Bax的表达有关。
Objective To observe the effect of Jianpi Huatan decoction on gene expression of P53,Bcl-2,Survivin and Bax in MFC tumor tissue and discuss the molecular mechanisms of its antitumor. Methods MFC cells inoculation in nude mice spleen,establishing nude mice liver metastasis model,which are divided into five groups randomly. After 4 weeks,detection the expression of P53,Bcl-2,Survivin and Bax protein and mRNA with immune histochemical method and RT-PCR method. Results Jianpi Huatan decoction can lower the expression of P53,Bcl-2 and Survivin protein and mRNA,raise the expression of Bax protein and mRNA. Conclusion Jianpi Huatan decoction resisting gastric cancer related to the cut of the expression of P53,Bcl-2,Survivin and the raise of the expression of Bax.
