摘要
目的探讨黄芪总苷对微波辐射致大鼠精子损伤的治疗作用。方法 75只二级Wistar雄性大鼠,随机分为正常对照组、辐射对照组、40 mg/(kg·d)黄芪总苷组、80 mg/(kg·d)黄芪总苷和160 mg/(kg·d)黄芪总苷组,每组15只。采用30 mW/cm~2微波全身辐射大鼠15 min,于辐射后当天开始灌胃给药,各组给药量分别为40 mg/(kg·d)、80 mg/(kg·d)和160 mg/(kg·d),正常对照组和辐射对照组均给予等比体积的蒸馏水,每天1次,连续14 d。分别于给药7 d(辐射后7 d)、停药后6 h(辐射后14 d)、停药后7 d(辐射后21 d)处死大鼠,采用SCA精子动态分析系统检测大鼠精子活动度、活力参数、运动参数,HE染色观测精子畸形率。结果 30 mW/cm~2微波辐射后7 d、14 d、21 d,大鼠精子活动度、活力参数、运动参数下降,畸形率升高(P<0.05或P<0.01);给药7 d,停药后6 h、7 d,80~160 mg/(kg·d)黄芪总苷组与辐射对照组比较,大鼠精子活动度、活力参数、运动参数升高,畸形率下降(P<0.05或P<0.01)。结论 30 mW/cm~2微波辐射可引起大鼠精子损伤,表现为大鼠精子活动度、活力参数、运动参数下降,畸形率升高;80和160 mg/(kg·d)黄芪总苷具有明显治疗微波辐射致精子损伤的作用,有效剂量为80 mg/(kg·d)。
        Objective To study the effect of astragaloside on sperm injury in rats induced by microwaveradiation. Methods Seventy five male Wistar rats were divided into 5 groups randomly: normal control(15 rats),microwave radiation control(15 rats),40 mg/(kg·d) astragaloside(15 rats),80 mg/(kg·d)astragaloside(15 rats) and 160 mg/(kg·d) astragaloside(15 rats) groups. The way of radiation was an average power density of 30 mW/cm~2 microwave radiation source with general uniform radiation 15 min.The rats received a daily intragastric administration from the day of radiation to 14 days after radiation once a day. The rats in each group were executed at 7 days of administration(7 days after radiation),6hours after drug withdrawal(14 days after radiation),7 days after drug withdrawal(21 days after radiation). The rat's sperm activate rate,motility parameters and motion parameters were detected by SCA sperm image analysis system. The malformation rate of sperms was calculated through H&E staining.Results The sperm activate rate,motility and motion parameters of rats were decreased(P < 0. 05 or P< 0. 01) and the sperm abnormality increased(P < 0. 05) at 7,14,21 day after 30 mW/cm~2 microwave radiation. The rat's sperm activate rate,motility and motion parameters were increased in 80 and 160 mg/(kg·d)astragaloside group compared to the radiation group. The ratio of sperm abnormality in 80 and 160mg/kg/d astragaloside groups decline D at 7 days of administration,6 hour and 7 days after the last intragastric administration(P < 0. 05 or P < 0. 01). Conclusions Microwave radiation at the average power densities of 30 mW/cm~2 could damage rats' sperms,such as the reduction of sperm motility,sperm motility parameters,sperm motion parameters and the increase of sperm abnormality; 80 ~ 160 mg/(kg · d)astragaloside groups exert therapeutic effect on injuries induced by microwave radiation. The effective dose of astragaloside on treating sperm injury induced by microwave radiation was 80 mg/(kg·d) astragaloside groups.
