摘要
目的探讨柴胡加龙骨牡蛎汤对更年期大鼠雌二醇和单胺类神经递质水平的影响。方法选取清洁级健康成年雌性SD大鼠24只,按照随机数字法分为对照组、柴胡加龙骨牡蛎汤干预组(CLM组)、柴胡加龙骨牡蛎汤+MEK激酶抑制剂组(CLM-PD组),每组8只。3组大鼠在第2天开始灌胃给药,CLM组及CLM-PD组予柴胡加龙骨牡蛎汤18. 4 g/kg,对照组予等量生理盐水,CLM-PD组同时口服10 mg/kg帕博西利布,持续给药至第7天。于第7天收集大鼠血清及脑垂体。检测血清雌二醇水平,脑垂体单胺类神经递质多巴胺(DA)、5-羟色胺(5-HT)和去甲肾上腺素(NE)水平,MEK与ERK1蛋白质表达水平。结果与对照组比较,CLM组与CLM-PD组大鼠血清雌激素水平均明显升高(均P<0. 05); CLM组与CLM-PD组大鼠血清雌激素水平比较差异无统计学意义(P>0. 05)。与对照组比较,CLM组大鼠脑垂体DA、5-HT和NE水平均明显升高(均P<0. 05),CLM-PD组大鼠脑垂体DA、5-HT和NE水平无显著变化(P>0. 05);与CLM组比较,CLM-PD组大鼠脑垂体DA、5-HT和NE水平均显著降低(P<0. 05)。与对照组比较,CLM组与CLM-PD组大鼠脑垂体MEK与ERK1蛋白表达水平均明显上调(均P<0. 05);与CLM组比较,CLM-PD组大鼠脑垂体MEK与ERK1蛋白表达水平明显下调(P<0. 05)。结论柴胡加龙骨牡蛎汤能提高更年期大鼠血清雌二醇水平,活化脑垂体内MEK/ERK信号通路和提高脑内单胺类神经递质DA、5-HT和NE的表达水平。
