摘要
目的:研究绞股蓝总黄酮抗心肌缺血保护作用及其抗氧化应激反应研究。方法:40只大鼠随机分为假手术组、模型组、绞股蓝总黄酮组、生理盐水组,每组10只,假手术组只穿线不结扎,其他组大鼠进行心肌缺血再灌注损伤造模。造模24 h后,测定各组大鼠心肌梗死面积;测定血清心肌酶(LDH、CK、GOT)活力、氧化应激指标(MDA、SOD、GSH-Px和CAT)表达水平;测定心肌组织中氧化应激指标(MDA、SOD、GSH-Px和CAT)表达水平。结果:与假手术组相比,模型组心肌梗死面积显著增大(P <0. 05),血清LDH、CK、GOT活力显著升高(P <0. 05),血清和心肌组织中MDA水平显著升高(P <0. 01),SOD、CAT和GSH-Px活力显著下降(P <0. 01)。与模型组和生理盐水组相比,绞股蓝总黄酮组心肌梗死面积明显减小(P <0. 05),血清LDH、CK、GOT活力显著升高(P <0. 05),血清和心肌组织中MDA水平显著升高(P <0. 01),SOD、CAT和GSH-Px活力显著下降(P <0. 01)。结论:绞股蓝总黄酮可以显著改善缺血再灌注大鼠氧化应激反应,减少心肌梗死面积,起到保护心肌作用。
Objective:To study the protective effect of total flavones from Gynura divaricata against myocardial ischemia and its anti-oxidative stress response. Methods: Forty rats were randomly divided into sham operation group,model group,total gypenosides group and normal saline group,with 10 rats in each group. Sham-operated rats were not ligated by threading. The other rats were subjected to myocardial ischemia-reperfusion injury model. After 24 h of modeling,myocardial infarct size was determined in each group. The activities of serum myocardial enzymes( LDH,CK,GOT) and the expression of oxidative stress indicators( MDA,SOD,GSH-Px and CAT) expression levels were compared. Results:Compared with the sham operation group,the area ofmyocardial infarction in model group was significantly increased( P <0. 05);the activities of serum LDH,CK and GOT were significantly increased( P < 0. 05) and the levels of MDA in serum and myocardial tissue were significantly increased( P < 0. 01). The activities of SOD,CAT and GSH-Px were significantly decreased( P < 0. 01). Compared with the model group and the saline group,the myocardial infarct size in the total flavonoids of Gynostemma pentaphyllum decreased significantly( P < 0. 05); the activities of serum LDH,CK and GOT increased significantly( P < 0. 05) and MDA levels in serum and myocardium significantly increased( P < 0. 01).The activities of SOD,CAT and GSH-Px decreased significantly( P < 0. 01). Conclusion: The total flavonoids of Gynostemma can significantly improve the oxidative stress response of rats with ischemia-reperfusion,reduce the area of myocardial infarction and plays a role in protecting myocardium.
