摘要
目的 研究黄连与吴茱萸不同组分配伍的抗氧化性和抗菌活性差异,为黄连与吴茱萸配伍系列经典类方的临床应用提供依据。方法 水提法制备黄连与吴茱萸经典配伍系列(左金丸、甘露散、茱萸丸、反左金丸),通过抑菌圈法测定不同配伍系列对大肠杆菌和金黄色葡萄球菌的抑菌活性,通过改良微量二倍稀释法测定不同配伍系列的最小抑菌浓度(MIC)。结果 4种配伍方剂对金黄色葡萄球菌有很强的抑菌作用,对大肠杆菌的抑菌作用较弱,抑菌效果顺序:左金丸>甘露散>茱萸丸>反左金丸,并推测抑菌效果与黄连总碱含量降低相关。以Vc作为阳性对照,采用1,1-二苯基-2-苦基肼(DPPH·)自由基、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS~+·)自由基清除试验对比各配伍系列对自由基清除活性。结果表明抗氧化作用大小为:反左金丸>茱萸丸>左金丸>甘露散,并推测抗氧化作用的物质基础可能是总酚含量。结论 黄连与吴茱萸经典配伍系列中左金丸具有较强的抗菌活性,反左金丸的抗氧化性最强,差异机制可能与配伍比例改变后活性物质含量差异相关。
        Objective This study compared the antibacterial and antioxidant activities of similar prescriptions of Coptis chinensis and Evodiae rutaecarpa to provide basis for clinical application of similar prescriptions. Methods The antibacterial activity against three bacteria Escherichia coli, and Staphylococcus aureus of the aqueous extract from similar prescriptions(Zuojinwan, Ganlusan, Zhuyuwan and Anti-Zuojinwan) were determined by bacteriostatic ring method,and the minimum inhibitory concentration(MIC)was detected with by microdilution method. Results Zuojinwan and its prescriptions had strong bacteriostatic effect on Staphylococcus aureus, but weak bacteriostatic effect on Escherichia coli. The order of inhibition circle size was: Zuojinwan > Ganlusan > Zhuyuwan > Anti-Zuojinwan. With Vc as the positive control, 1-bis-2-phenyl picrylhydrazine( DPPH) radical and 2,2-azino-two( 3-ethyl-benzothiazole-6-sulfonic acid) two ammonium salt( ABTS) were used to assay the free radical scavenging activity of match-pair of Coptis-Evodia. The antioxidation effect of similar prescriptions of Coptis chinensis and Evodiae rutaecarpa was as follows: Anti-Zuojinwan > Zhuyuwan > Zuojinwan > Ganlusan. Conclusion oth the series of similar prescriptions of compatibility have antioxidant and antibacterial activities, while Zuojinwan has the storngest antibacterial effect and Anti-Zuojinwan has the storngest antioxidant activities.
