摘要
目的优化芪黄扶正颗粒的提取纯化工艺。方法以马钱苷、芍药苷、阿魏酸提取率、指纹图谱特征峰峰面积、干浸膏得率等为评价指标,考察乙醇浓度、药液比、提取时间、提取次数对芪黄扶正颗粒提取工艺的影响;以马钱苷、芍药苷、阿魏酸转移率、指纹图谱特征峰峰面积、干浸膏得率等为综合评价指标,考察加样顺序、药液乙醇含量、药液浓度、絮凝剂用量对芪黄扶正颗粒纯化工艺的影响。结果最佳提取工艺为:乙醇浓度60%,乙醇8倍用量,每次提取1.5h,提取2次;最佳纯化工艺为:絮凝剂中B组分先加,A组分后加,药液乙醇含量30%,药液浓度0.1409g·ml~(-1),絮凝剂中B组分用量4%,A组分2%。结论优化的提取纯化工艺稳定、可靠,为获得有效和低给药剂量的芪黄扶正颗粒剂奠定了试验基础。
        Objective To optimize the extraction and purification process of Qihuang Fuzhengparticles.Methods The concentration and amount of ethanol,extraction time,and the number of times of extraction were investigated to optimize the extraction process of Qihuang Fuzhengparticles with the extraction rate of loganin,paeoniflorin,and ferulic acid,dry extract yield,and fingerprint characteristic peak area score as evaluation indexes.The effect of the order in which samples were added,ethanol concentration in the concentrated solution,concentration of extracting solution,and flocculant agent amount on the purification process was also investigated.Results The best extraction technology was optimized as follows:60% ethanol,eight times the ethanol volume,extraction twice,each time for 1.5 h.The best purification technology was optimized as follows:flocculant agent B was added before A agent,30% ethanol in the concentrated solution,0.1409 g·ml~(-1) of the concentration of the extraction liquid,4% flocculant agent B and 2% agent A.Conclusion The optimized extraction and purification process of Qihuang Fuzheng particles is stable and reliable,which will help produce effective and low-dosage Qihuang Fuzhengparticles.
