复元活血汤对大鼠股骨骨折后血清生长激素及骨痂GH受体表达水平影响

摘要
目的:探讨复元活血汤对大鼠股骨骨折后血清生长激素(GH)及骨痂GH受体表达水平影响。方法:选取由本院实验动物中心提供清洁级雄性SD大鼠30只,大鼠腹腔内注射10%水合氯醛麻醉,股骨中断使用低速牙科钻将股骨切断,使用克氏针由髓腔内逆行固定造模,观察组大鼠给予复元活血汤灌胃,对照组则使用等剂量生理盐水灌胃,每天一次,共进行28天,在给药后10天、20天、28天分别选取5只大鼠,麻醉处死,全自动血流变测试仪器检测给药10、20、28天后不同剪切速率时大鼠血浆粘度与全血粘度,比色法检测血清磷(P)、钙(Ca)、ALP含量,ELISA检测血清GH含量,免疫组法检测骨痂组织内GHR表达状况。结果:给药后20、28天观察组大鼠全血30s-1、5s-1及1s-1切变率均低于对照组,差异有统计学意义(P<0. 05);给药后20、28天观察组大鼠血清GH、P、Ca及ALP水平均高于对照组,差异有统计学意义(P<0. 05);给药20、28天后观察组大鼠GHR表达高于对照组,血浆粘度低于对照组,差异均有统计学意义(P<0. 05)。结论:复元活血汤可使股骨骨折大鼠血清GH活性增强,降解速度减缓,骨痂组织内GHR表达量升高,进而发挥加速骨折愈合作用。
        Objective: To investigate the effect of Fuyuan Huoxue Decoction on serum GH and callus GH receptor expression in femoral fractures of rats. Methods: Thirty male SD rats were randomly divided into experimental group and controlled group.Rats were anesthetized by intraperitoneal injection of 10% chloral hydrate,the femur was cut off with low-speed dental drill,and the Kirschner wire was retrograde fxed modeling,the observation group rats were given Fuyuan Huoxue Decoction gavage,the controlled group was treated with an equal dose of saline,once a day for a total of 28 days,10days after administration,20 days,28days were selected 5rats were anesthetized and sacrificed. The automatic blood rheology testing instrument was used to detect the plasma viscosity and whole blood viscosity of rats at different shear rates 10,20 and 28days after the administration. The contents of serum phosphorus(P),calcium(Ca),ALP content,serum GH levels by ELISA,immunohistochemistry method to detect the expression of GHR in callus tissue. Results: The shear rates of 30s-1,5s-1and 1s-1in the whole blood of the observation group were lower than those of the controlled group on the days 20 and 28after the administration,the difference was statistically significant(P<0. 05),and the serum GH,P,Ca and ALP levels in the 28-day of the observation group were higher than those in the controlled group(P<0. 05). After 20 and 28days,the GHR expression in the observation group was higher than that in the controlled group,plasma viscosity was lower than the controlled group,the difference was statistically significant(P<0. 05). Conclusion: Fuyuan Huoxue Decoction can increase the serum GH activity,reduce the degradation rate and increase the expression of GHR in callus tissue of rats with femoral fractures,and then accelerate the fracture healing.
