SCSA与SCD检测精子DNA碎片指数的比较分析

摘要
目的比较分析精子染色质结构测定法(SCSA)和精子染色质扩散法(SCD)两种方法在检测精子DNA碎片指数(DFI)时的结果。方法收集2017年5月至2018年4月前来我院就诊的男性患者精液标本165份,记录其基本信息,分别采用SCSA和SCD两种方法检测其DFI并进行比较分析。结果 SCSA与SCD检测DFI的结果显著相关(r=0.598,P<0.01);测得的DFI结果,SCD组(19.17±0.93)%高于SCSA组(13.64±0.81)%,差异具有统计学意义(P<0.01)。在重度低浓度组中,SCSA与SCD之间的差异无统计学意义(P>0.05)且不存在相关性(r=0.06, P>0.05)。结论 SCSA与SCD检测精子DFI的结果具有显著正相关,但SCD检测精子DFI的结果高于SCSA,临床诊疗中医生应根据两种检测方法的特点对DFI的检测结果进行综合判断。对于临床上重度低浓度精子的患者,其DFI结果的准确性要谨慎对待。
Objective To comparatively analyze the difference of two methods of sperm chromatin structure assay(SCSA) and sperm chromatin dispersion(SCD) in detecting sperm DNA fragment index(DFI). Methods 165 semen samples of male patients from May 2017 to April 2018 were collected and basic information of the patients was recorded. DFI was detected by SCSA and SCD respectively. Results The results of sperm DFI of SCSA and SCD were significantly correlated(r = 0.598, P <0.01), but DFI results of the SCD group(19.17±0.93)% were higher than those of the SCSA group(13.64±0.81)%. The difference was statistically significant(P < 0.01). In severe low concentration group, there was no significant difference in DFI results between SCSA group and SCD group(P > 0.05), and there was no correlation(r = 0.06, P >0.05). Conclusion The results of sperm DFI in SCSA group and SCD group have a significant positive correlation, but the results of sperm DFI in SCD group is higher than that in SCSA group. Doctors should comprehensively evaluate the results of DFI based on the characteristics of two methods in clinical diagnosis and treatment. For patients with clinically severe low concentration sperm, the accuracy of DFI results should be treated with caution.
