摘要
目的:观察榄香烯注射液对人肝癌HepG2细胞株体外增殖的影响及其特点,并初步探讨其可能的相关作用机制。方法:以5-氟尿嘧啶(5-FU)为阳性对照组,应用噻唑蓝比色法检测不同浓度榄香烯实验组HepG2细胞的吸光度值、增殖抑制率,并应用流式细胞检测技术(FCM)测定HepG2细胞周期分布特点。结果:HepG2细胞经榄香烯作用36 h后折光率减弱,细胞体积缩小、漂浮细胞增多,细胞生长受到显著抑制,并呈现出明显的时间依赖效应关系。在细胞周期分布特点及抑制率参数上,榄香烯组与空白对照组相比有统计学差异(P<0. 01),而与5-Fu组比较,二者之间无统计学差异(P>0. 05)。同时榄香烯使HepG2细胞周期分布特点发生改变,表现为G0/G1期和G2/M期细胞比例上升,S期细胞比例下降,并且与5-Fu组呈现出相似的趋势。结论:榄香烯注射液在体外能通过抑制肝癌HepG2细胞株DNA复制发挥抑制其增殖的作用。
        Objective: To explore the effects of Elemene on the proliferation of human hepatocellular cancer(HCC) cell HepG2 in vitro, and to discuss the possible molecular mechanism.Methods: 5-fluorouracil(5-Fu) group was set as positive control, the absorption value and cell inhibition rate of proliferation of HepG2 cells were detected by MTT test, and characteristics of cell cycle distribution were studied by FCM after treatment by various concentration of Elemene.Results: Refractive index of HepG2 cells was decreased and they shrank gradually along with the increase of concentration, while dead cells were increased. The proliferation of HepG2 cells was inhibited markedly in a time-dependent manner. There were significant differences in the parameters of cell cycle distribution and inhibition rates between Elemene group and negative control group(P<0. 01); however, no statistical significance was observed between elemene group and 5-Fu group(P>0. 05). The characteristics of cell cycle distribution changed dramatically when HepG2 cells were cultured with Elemene in vitro: the percentage of cells in stage G0/G1 and G2/M was increased, while that of cells in stage S reduced; this trend was similar to that of 5-Fu group.Conclusion: Elemene can effectively inhibit the proliferation of Hep G2 in vitro via inhibiting DNA synthesis.
