摘要
本研究从亚洲百合(Lilium Asiatic hybrids)花粉败育品种‘小小的吻’中克隆获得一个MYB转录因子基因LaMYB26。序列分析显示,LaMYB26基因的开放阅读框(ORF)长762 bp,编码253个氨基酸。结构域分析显示,LaMYB26含有2个典型的SANT结构域,属于MYB转录因子家族中的R2R3-MYB型。多序列比对发现,LaMYB26与其他物种的MYB类蛋白的氨基酸序列有57.78%的相似性。理化性质、亲/疏水性和无序化分析显示, LaMYB26转录因子是亲水性蛋白且偏碱性,存在无序化区域。信号肽和跨膜结构域分析表明,其不存在信号肽和跨膜结构域。空间结构分析显示, LaMYB26有2个典型的α-螺旋串联。实时荧光定量PCR (qRT-PCR)显示, LaMYB26基因在败育时期表达量明显上调,且在花药中特异表达。结果显示, LaMYB26基因在百合花粉败育过程中可能发挥着重要作用,对LaMYB26的生物学信息学分析拟为深入研究其功能及作用机制提供参考。
        LaMYB26, a MYB transcription factor, was cloned from lily(Lilium Asiatic hybrids) cultivar ‘Little Kiss’ with pollen abortion in this study. Sequence analysis revealed that the open reading frame of LaMYB26 gene is 762 bp and encodes 253 amino acids. LaMYB26 transcription factor contains two typical SANT domains and belongs to the R2 R3-MYB type in the MYB family transcription factor. Multi-sequence comparison founds that the amino acid sequence of LaMYB26 is 57.78% in similarity to those of MYB proteins in other species. The physicochemical properties, hydrophilicity/hydrophobicity and disorder analyses of LaMYB26 transcription factor demonstrates that it is a hydrophilic and alkaline protein with a disordered region. Analyses of signal peptide, transmembrane domain, and spatial structure indicate that LaMYB26 has no signal peptide and transmembrane domain, but has two typical α-helix series. Quantitative real-time PCR showed that the expression level of LaMYB26 gene was up-regulated during pollen abortion stage and was specifically expressed in lily anther. Taken together, the results suggest that LaMYB26 gene may play an important role in lily pollen abortion, and the current bioinformatic analyses of LaMYB26 would lay a foundation for further studies of its function and mechanism.
