摘要
目的:采用高效液相色谱法同时测定广西区内不同产地楠藤中绿原酸、隐绿原酸、3,5-O-二咖啡酰基奎宁酸、3,4-O-二咖啡酰基奎宁酸及4,5-O-二咖啡酰基奎宁酸5种成分的含量。方法:色谱柱为Kromasil C18(4.6mm×250mm,5μm);以乙腈(A)-0.4%磷酸水溶液(B)为流动相,梯度洗脱:0~15 min,89%B;15~25 min,89%~80%B;25~50 min,80%B;流速1.0mL·min-1;检测波长328nm;柱温30℃。结果:5种成分在各自进行范围内均呈良好的线性关系,平均加样回收率在98.92%~102.71%之间;广西区内不同产地的楠藤样品中5种成分的总含量在0.418%~1.580%,不同地区的总含量有一定的差异。结论:该高效液相色谱法快速、准确,可用于楠藤中5种咖啡酰基奎宁酸类成分的含量测定;楠藤中该5种成分含量具在一定的地域差异性。
Objective:To develop an HPLC method for the simultaneous determination of five components,including chlorogenic acid,cyrptochlorogenic acid,3,4-O-dicaffeoylquinic acid,3,5-O-dicaffeoylquinic acid and 4,5-O-dicaffeoylquinic acid,in Mussaenda erosa Champ.from different districts in Guangxi.Methods:The separation was completed on a Kromasil C18(4.6 mm×250 mm,5μm)column.The mobile phase was acetonitrile-0.4%phosphoric acid with the gradient elution:0-15 min,89% B;15-25 min,89%-80% B;25-50 min,80% B.The flow rate was 1.0 mLmin-1,the detection wavelength was 328 nm and the column temperature was 30℃.Results:The standard curve revealed a good linear relationship for this five components and the average recovers were between 98.92%to 102.71%.The total content of the five components in the Nantong samples from different regions in Guangxi was 0.418%to 1.580%,and the total content in different regions had a certain difference.Conclusion:The developed method can be used for the rapid and accurate determination of five components in Mussaenda erosa Champ.
