摘要
目的筛选山楂叶多糖的最佳提取条件及抗氧化性研究。方法采用乙醇溶液做提取试剂,超声辅助提取山楂叶多糖,通过单因素、正交试验确定最优提取条件,通过Sevage法除去其中蛋白质,用1,1-二苯基-2-三硝基苯肼自由基(DPPH·)法测定提取物即多糖的自由基清除率。结果最佳提取条件为料液比为1∶20,60℃提取20 min,提取2次;山楂叶多糖对1,1-二苯基-2-三硝基苯肼自由基清除率达到65.1%。结论超声辅助可以较好提取山楂叶多糖,所提取的山楂叶多糖具有良好的抗氧化活性。
        Objective To screen the optimum extraction conditions and antioxidative activity of polysaccharide from hawthorn leaves.Methods Ethanol extract was used as an extraction reagent and ultrasonic assisted extraction of polysaccharides from hawthorn leaves. The optimal extraction conditions were determined by single factor,orthogonal test,and protein was removed by Sevage method.The radical scavenging rate of polysaccharides was determined by DPPH·.Results The optimum extraction conditions were as follows: the ratio of material to liquid is 1 ∶ 20,extracted at 60 ℃ for 20 min,and 2 times were extracted.The scavenging rate of hawthorn leaf polysaccharide on DPPH· was 65.1%. Conclusion The polysaccharide of hawthorn leaves can be extracted with the aid of ultrasound,and the extracted polysaccharides from hawthorn leaves have good antioxidant activity.
