摘要
目的:探讨中药大黄提取物芦荟大黄素对培养人成纤维细胞Smad 2、Ⅰ型胶原蛋白表达及细胞周期的影响,分析芦荟大黄素对成纤维细胞的调控作用及作用机制。方法:体外培养人成纤维细胞,用芦荟大黄素进行干扰,分为对照组(CG)、低剂量组(50-TG)、中剂量组(100-TG)、高剂量组(500-TG),观察细胞生长特征,并用量子点免疫荧光技术检测Smad 2、Ⅰ型胶原的表达,流式细胞仪检测的细胞周期。结果:(1)干扰48h后,实验组Smad 2表达的荧光强度,与对照组之间差异有统计学意义(P <0. 05);(2)实验组Ⅰ型胶原表达,与对照组之间差异有统计学意义(P <0. 05);(3)流式细胞仪检测细胞周期结果显示,对照组、实验组G1期细胞所占比例比较,差异有统计学意义(P <0. 05)。结论:芦荟大黄素对培养成纤维细胞的Smad 2、Ⅰ型胶原蛋白表达具有调控作用,并能影响其细胞周期和细胞的活性,可能是含有芦荟大黄素的中药如芦荟、大黄等调控成纤维细胞功能的主要机制之一。
Objective: To explore the effect of the aloe-emodin from traditional Chinese medicine on the expression of Smad2,collagenⅠin fibroblasts and cell cycle of fibroblasts from human,and study role of adjustment and the mechanism of aloeemodin on fibroblasts in vitro. Methods: Culture human fibroblasts in vitro and interfere the cells with aloe-emodin,then detect the expression of Smad 2,collagenⅠwith immunofluorescent,and cell cycle with flow cytometry( FCM). Results:( 1) After 48 hours,there is significant difference of immunofluorescent of the expression of Smad 2 between the control group and the treatment group( P < 0. 05);( 2) after 48 hours,there is significant difference of immunofluorescent of the expression of collagenⅠ between the control group and the treatment group( P < 0. 05);( 3) there is significant difference of the rate of G1 cells between the control group and the treatment group( P < 0. 05). Conclusion: Aloe-emodin have the role of adjustment the cell cycle and expression of collagen Ⅰ,collagen Ⅲ in the fibroblasts in vitro,that may be the main mechanisms of the aloe-emodin from traditional Chinese medicine to regulating the function of fibroblasts.
