摘要
目的观察右美托咪定预处理对盲肠结扎穿孔(CLP)脓毒症模型大鼠急性肾损伤的影响及TLR4表达变化。方法 24只大鼠随机分为三组,假手术(SHAM)组,模型(CLP)组和右美托咪定(DEX)组,每组8只,各组在造模前30min分别腹腔注射等剂量生理盐水或右美托咪定100μg·kg~(-1),随后行CLP造模并观察24h,酶联免疫吸附法测定血清TNF-α和IL-6,HE染色观察肾组织病理结构变化,荧光定量PCR测定肾组织TLR4 mRNA表达。结果与CLP组比较,DEX组血清TNF-α和IL-6含量明显减少(P<0.01),肾组织的损伤情况明显减轻,TLR4 mRNA表达亦显著减少(P<0.01)。结论右美托咪定预处理能减轻脓毒症引起的肾组织炎症反应,具有一定保护作用。
        OBJECTIVE To observe the effects of dexmedetomidine preconditioning on renal acute injury and the TLR4 expression changes in rats with cecal ligation perforation( CLP). METHODS 24 rats were randomly divided into three groups,SHAM group,model( CLP) group and dexmedetomidine( DEX) group,each group includes eight rats,and recerved isodose physiological saline or dexmedetomidine( 100μg·kg-1) by intraperitoneal respectively 30 minutes before making model,then makes the CLP building 24 h and observes it. TNF-α and IL-6 in the serum were measured by ELISA. The changes of pathological structure of renal tissue were observed with hematoxylin-eosin staining and the expression of renal TLR4 mR NA was measured by RT PCR. RESULTS Compared with group CLP,the expression of TNF-α and IL-6 in the serum significantly decreased( P < 0. 01),the change of kidney his-tological showed lessened injury,the level of TLR4 mR NA was lower apparently( P < 0. 01) in group DEX. CONCLUSION The pretreatment of dexmedetomidine can reduce the inflammatory reaction of nephridial tissue caused by sepsis and have a certain protective effect.
