加味过敏煎不同拆方组分对膜性肾病大鼠足细胞裂孔膜蛋白Nephrin的影响

摘要
目的:观察加味过敏煎不同拆方组分对膜性肾病大鼠肾组织Nephrin表达的影响。方法:采用阳离子化牛血清白蛋白尾静脉注射的方法诱导膜性肾病大鼠模型、将加味过敏煎按调理开合、健脾益肾、活血化瘀拆方成不同组分对其进行灌胃干预,检测24h尿蛋白定量、肾组织病理、Nephrin蛋白及m RNA表达。结果:加味过敏煎组及其拆方组大鼠24h尿蛋白定量及肾脏病理损伤明显缓解(P<0.05)。加味过敏煎组及其拆方组大鼠肾小球Nephrin蛋白及m RNA表达量明显增多(P<0.05),其中去健脾益肾组、活血化瘀组和去调理开合组改善效果较好(P<0.05)。结论:活血化瘀组分在膜性肾病的尿蛋白恢复方面具有重要的作用,且能促进Nephrin蛋白及基因的表达,可能是蛋白尿恢复的机制之一。
Objective: To observe the effects of Jiawei Guomin Decoction and its disassembled prescription on protein expression of Nephrin in the podocyte of membranous nephropathy rats.Methods: The model rats with membranous nephropathy was established by injecting vein with cationic bovine serum protein(C-BSA),which were treated by the different decomposed Jiawei Guomin Decoction that were divided according to their functions of regulating opening and closing of sweat pores(A),invigorating spleen and kidney(B),promoting blood circulation to remove stasis(C),and the 24 hour urinary protein,expression of Nephrin protein and m RNA in kidney tissue and the pathology of kidney tissue were respectively detected.Results: The 24 hour urinary protein and pathology of kidney tissue of Jiawei Guomin group and its Disassembled prescription groups were significantly relieved(P<0.05).The protein and m RNA expression of Nephrin Jiawei Guomin group and its ingredients groups were significantly increased(P<0.05),and AC,C and BC group had better effect.Conclusion: Huoxue Huayu ingredients plays an important role in the recovery of urinary protein in membranous nephropathy and can promote the protein and gene expression of Nephrin,which may be one of the mechanisms of proteinuria recovery.
