Galectin-3基因调控Wnt/β-catenin信号通路对肺癌细胞凋亡的影响

摘要
目的探讨半乳糖凝集素(Galectin)-3基因对肺癌细胞凋亡的影响及机制。方法将NC-siRNA、Galectin-3-siRNA1和Galectin-3-siRNA2转染人肺癌A549细胞,未转染任何的siRNA作为对照组,转染48 h后提取细胞中的蛋白,Western印迹检测各组细胞中Galectin-3蛋白表达;流式细胞仪检测细胞凋亡率;Western印迹检测酶切含半胱氨酸天冬氨酸蛋白水解酶(Cleaved caspase)3、β-链蛋白(catenin)、细胞周期蛋白(Cyclin)D1蛋白表达。结果 NC-siRNA组Galectin-3蛋白表达与对照组差异无统计学意义(P>0.05),Galectin-3-siRNA1、Galectin-3-siRNA2组Galectin-3蛋白表达均显著低于对照组(P<0.01),Galectin-3-siRNA1组的抑制效果更明显,选择作为后续研究;与对照组及NC-siRNA组比较,Galectin-3-siRNA组β-catenin、Cyclin D1蛋白表达显著降低,细胞凋亡率及Cleaved caspase3蛋白表达显著升高(P<0.01)。结论抑制Galectin-3在肺癌细胞中的表达可诱导细胞凋亡,其机制与抑制Wnt/β-catenin信号通路有关。
