美洲大蠊粉对腹泻小鼠结肠酶活性及其体内菌群增殖的影响

摘要
为了明确美洲大蠊粉对腹泻小鼠结肠酶系活性及菌落数的影响,采用腹腔注射大肠杆菌建立小鼠腹泻模型,以饲料添加含量为8%(高剂量组)、5%(中剂量组)、2.5%(低剂量组)的美洲大蠊粉对其进行治疗,以紫外分光光度法检测小鼠结肠中SOD、POD、CAT、GSH、ACHE、ACP、LDH、AMS、LPS活性,以平板菌落计数法统计小鼠结肠的总菌和大肠杆菌数量。结果显示,腹泻引起小鼠结肠组织中相关酶系活性下降。饲喂含不同剂量的美洲大蠊粉的饲料后,高剂量治疗组小鼠结肠的SOD、高剂量和中剂量治疗组小鼠的POD、3种剂量治疗组小鼠的CAT均显著高于模型组(p<0.05);各剂量治疗组小鼠的GSH、ACHE显著高于模型组(p<0.05);高剂量和中剂量组小鼠的ACP显著高于模型组(p<0.05);各剂量治疗组的LPS显著高于模型组(p<0.05);高剂量和中剂量治疗组的AMS、LDH显著高于模型组(p<0.05);各剂量组小鼠的大肠杆菌数、总菌数显著低于模型组(p<0.05),其中高剂量治疗组小鼠的大肠杆菌数量低于空白组。上述结果表明,美洲大蠊粉能够增强小鼠抗腹泻能力。且以高、中剂量治疗腹泻小鼠的效果最好。
        In order to clarify the effect of Periplaneta americana powder on the activity of enzyme system and colony number in diarrhea mouse of colon. The diarrhea mouse model was established by intraperitoneal injection with Escherichia coli, and the diarrhea mice were treated by the P.americana powder with 8%(high dose group), 5%(medium dose group), and 2.5%(low dose group) supplemented in daily feed. The activity of SOD, POD, CAT, GSH, ACHE, ACP, LDH, AMS and LPS in the mouse colon were detected by ultraviolet spectrophotometry, and total number of bacteria and E.coli in mouse colon were counted by plate colony counting method. The results showed that diarrhea caused a decrease of the activity of related enzyme system in the colon tissue of mouse. After the mice were treated with P.americana powder, the SOD of the mice in the high dose group, the POD in the high and middle dose group, as well as the CAT in all of three dose groups were significantly higher than that in the model group; the GSH and ACHE of mouse in each dose treatment group were significantly higher than that of the model group, while the ACP in the high and middle dose group was significantly higher than that of the model group; the LPS in each dose treatment group was significantly higher than that in model group, while the AMS, and LPS of the high and medium dose treatment group were significantly higher than that in the model group; the number of E.coli and total bacteria in each dose group were significantly lower than that in the model in group, among which the number of E.coli in the high-dose treatment group was even lower than that in blank control group. In conclusion, the P.americana powder could enhance the ability of anti-diarrhea in mice.The effect of high and medium dose treatment was the best.
