摘要
采用光谱法、电化学循环伏安法及粘度法研究了大黄酸及大黄酸铜配合物与脱氧核糖核酸(DNA)的相互作用。研究发现DNA对大黄酸及大黄酸铜配合物的紫外光谱有减色作用,大黄酸及大黄酸铜配合物的加入对DNA-溴化乙啶体系的荧光有强的猝灭作用,且随DNA加入,大黄酸及大黄酸铜配合物的氧化还原峰均有降低。实验结果表明大黄酸及大黄酸铜配合物与DNA发生嵌插作用,形成了复合物,且大黄酸铜配合物与DNA的作用大于大黄酸。
        The interaction of the rhein and rhein-Cu(Ⅱ) complex with calf thymus DNA was studied by electronic absorption spectroscopy, fluorescence spectrometry, circular dichroism spectrometry, cyclic voltammetry and viscosity method. It was found that UV absorption spectrum showed that the absorption peak of rhein and rhein-Cu(Ⅱ) complex declined with the addition of DNA.The rhein and rhein-Cu complex can lead to the intrinsic fluorescence quenching of DNA-EB system through a static quenching procedure. The oxidation peak currents of rhein and rhein-Cu decreased and the peak potentials shifted positively with adding DNA. These results indicate that rhein and rhein-Cu complex can react with DNA mainly in a mode of intercalation. The binding force of rhein-Cu is stronger than rhein.
