摘要
目的:实验通过温阳化浊通络方含药血清处理皮肤成纤维细胞研究其抗纤维化机制,进而探讨其在治疗系统性硬化病纤维化的作用机制。方法:采用血清药理学方法以不同浓度的温阳化浊通络方含药血清与人系统性硬化病皮肤成纤维细胞共同孵育,并与空白血清对照。研究温阳化浊通络方含药血清对体外皮肤成纤维细胞中Wnt/β-catenin信号通路及纤维化相关标志物的影响。结果:与空白血清组相比,温阳化浊通络方含药血清能够显著抑制β-catenin、c-myc、Fn1、Collagen I和CTGF的mRNA和蛋白表达水平,以及TOP/FOP flash双荧光素酶报告基因活性,并且随着温阳化浊通络方含药血清浓度的提高,其抑制作用增强。更加重要的是,温阳化浊通络方含药血清能够抑制β-catenin/TCF4转录调控复合物的形成,其抑制作用与温阳化浊通络方含药血清浓度呈正相关。结论:温阳化浊通络方含药血清对体外皮肤成纤维细胞中Wnt/β-catenin信号通路及纤维化相关标志物有明显的抑制作用,为温阳化浊通络方抗系统性硬化病纤维化提供了科学依据。
        Objective:The study is to detect the anti-fibrosis mechanism by treating skin fibroblasts with medicated serum containing Wenyang Huazhuo Tongluo Recipe,and then explore its mechanism in the treatment of systemic sclerosis fibrosis.Methods:Human systemic sclerosis skin fibroblasts was incubated with medicated serum of different concentrations of Wenyang Huazhuo Tongluo Recipe.And studying the effect of medicated serum on Wnt/β-catenin signaling pathway and fibrosis-associated markers in human skin fibroblasts.Results:Compared with the blank serum group,medicated serum could significantly inhibit the mRNA and protein expression levels of β-catenin,c-myc,Fn1,Collagen I and CTGF as well as TOP/FOP flash Dual-luciferase reported gene activity and with the concentration of medicated serum increased and the inhibition was enhanced.More importantly,medicated serum could inhibit the formation of β-catenin/TCF4 transcriptional regulatory complex and its inhibitory effect was positively correlated with the concentration of medicated serum.Conclusion:Medicated serum containing Wenyang Huazhuo Tongluo Recipe inhibited Wnt/β-catenin signaling pathway and fibrosis-associated markers in human skin fibroblasts,providing a scientific basis for the prevention of systemic sclerosis fibrosis by Wenyang Huazhuo Tongluo Recipe.
