虎耳草有效成分的提取及其生物活性的研究

摘要
以虎耳草为原料,探讨了料液比、乙醇浓度、回流次数对虎耳草黄酮含量的影响.并测定了虎耳草提取液的抗氧化、抗菌及酶抑制性能.结果表明,反应料液比、乙醇体积分数、回流次数对黄酮含量有不同程度的影响,虎耳草黄酮的最佳提取条件为:料液比为1∶30,溶剂为体积分数90%乙醇水溶液,温度为100℃,回流浸提3次,黄酮含量最高达42.04 mg·g~(-1);虎耳草提取物对α-淀粉酶抑制作用效果最好的条件是质量浓度为8.0 mg·m L~(-1)、pH=4.5、温度为40℃;虎耳草提取物有较强的羟基自由基清除能力,清除率最高达97.57%;虎耳草对大肠杆菌及枯草芽孢杆菌没有抑制效果,虎耳草对青霉、曲霉均具有很好的抑制作用,并且对青霉的抑制作用大于曲霉,最大抑菌圈直径为20.06 mm.
        Saxifrage as raw material,effects of the ratio of material and liquid,the concentration of ethanol and the time of efluxing on the yield of total flavonoids were discussed in this paper.And the antioxidant,antibacterial,enzyme inhibition performance of saxifrage extract were also researched. The results showed that the ratio of material and liquid,the concentration of ethanol and the time of efluxing had obvious influence on the yield of total flavonoids. The optium conditions were the ratio of material and liquid 1∶ 30,90% ethanol aqueous solution,the temperature of 100℃,reflux extraction 3 times. The yield of total flavonoids was 42. 04 mg·g~(-1). The optium conditions of saxifrage extract on alpha amylase inhibition effect were the concentration of 8. 0 mg ·m L~(-1),pH = 4. 5,temperature of 40 ℃. Saxifrage extract had strong ability of scavenging hydroxyl free radical,the clearance rate was 97. 57%. Saxifrage extract had no inhibition effect on escherichia coli and bacillus subtilis. Saxifrage had a good inhibitory effect on penicillium and aspergillus,and the inhibition effect on penicillium was greater than aspergillus,maximum bacteriostatic circle diameter was 20. 06 mm.
