摘要
目的研究分级醇沉的柳茶多糖(SAPs)含量、单糖组成及抗氧化活性。方法采用水提、分级醇沉法制备SAPs,用硫酸-苯酚法测定多糖的含量,1-苯基-3-甲基-5-吡唑啉酮(PMP)衍生化HPLC法分析单糖的组成,红外分光光度法研究多糖的构型,DPPH·法评价分级多糖的抗氧化活性。结果分级醇沉SAPs含量的高低顺序为60%> 40%> 80%> 20%;葡萄糖、半乳糖和阿拉伯糖在各级醇沉SAPs三氟乙酸水解产物中的含量均最高,其次是半乳糖醛酸、葡萄糖醛酸、甘露糖和核糖,未检测到鼠李糖和木糖;分级醇沉SAPs的抗氧化性优劣顺序为40%> 80%> 60%> 20%,40%醇沉多糖浓度为0. 4 mg·m L~(-1)时对DPPH·清除能力与0. 04 mg·m L~(-1)抗坏血酸相当,其半数抑制浓度IC50=0. 235 mg·m L~(-1)。结论实验结果为柳茶多糖进一步化学、工艺和药理研究提供了参考。
        OBJECTIVE To study the content,monosaccharide composition and antioxidant activity of different Sibiraea angustata polysaccharides( SAPs) which were precipitated using gradient concentrations of ethanol. METHODS Different SAPs were extracted by water and precipitated with gradient concentrations of ethanol. The content of polysaccharides was determined by sulfuric acid-phenol method. The composition of monosaccharide was determined by HPLC with PMP derivatization. Infrared spectrophotometry was used to study the configuration of SAPs. The antioxidant activity of SAPs was evaluated by DPPH· method. RESULTS The order of the contents of different SAPs was 60% > 40% > 80% > 20%. The contents of glucose,galactose and arabinose were the highest in the TFA hydrolysis products of different SAPs,followed by galacturonic acid,glucuronic acid,mannose and ribose,but no rhamnose and xylose were detected. The order of antioxidant activity was 40% > 80% > 60% > 20%. The scavenging ability of 0. 4 mg·m L~(-1) SAPs which were precipitated by 40% ethanol to DPPH· was almost the same as that of 0. 04 mg·m L~(-1) Vit C,and its IC50 value was 0. 235 mg·m L~(-1). CONCLUSION The research provides references for further studies on chemical constituents,techniques and pharmacology of SAPs.
