摘要
为探讨大骨鸡遗传多样性和系统进化,了解大骨鸡母系起源,以大骨鸡为试验素材,通过PCR扩增和测序技术,对大骨鸡线粒体DNA (mtDNA)控制区全序列进行测序,并与GenBank公布的19条红色原鸡D-loop区全序列进行比对分析。结果表明:30只大骨鸡mtDNA控制区序列全长为1 231或1 232bp,这2个序列均有15只个体,但1 231bp序列的个体在859bp处存在C碱基缺失。30只个体共发现22个多态性位点、7种单倍型。系统发育分析显示,7种单倍型可分为A、B、C和E 4个分支。其中A和B分支均与原鸡滇南亚种聚为一类,推测可能起源于红色原鸡滇南亚种;C分支与4个原鸡亚种聚为一类,推测有多个母系起源;E分支与原鸡印度亚种聚为一类,推测可能起源于红色原鸡印度亚种。这一研究从分子水平上为大骨鸡遗传资源保护和开发利用提供了参考。
        To investigate the genetic diversity and phyletic evolution and understand the maternal origin of Dagu chicken,Dagu chickens were used as material,by PCR amplification and sequencing technique,complete mtDNA D-loop sequences of Dagu chickens were analyzed with the 19 red jungle fowl sequences in GenBank.The results showed that,the D-loop region of Dagu chicken was 1 231 or 1 232 bp,the 1 231 bp haplotype had a base Cdeficiency in 859 site.In the30 samples assayed,22 mutation sites and 7haplotypes were found.Phylogenetic analysis showed that 7haplotypes of Dagu chicken could be converged into four clades.The clade A and clade B were clustered with Gallus gallus spadiceus,indicating that two clades may originate from the continental subspecies of G.gallus spadiceus.The clade C was clustered with four subspecies of red jungle fowl,which may have multiple origins.The clade E was clustered with G.gallus murghi,that clade E was originated from the continental subspecies of G.gallus murghi.The results would be a reference for genetic resources conservation and development and utilization of Dagu chicken.
