摘要
目的:分析甘遂入药对比呋塞米对癌性腹水大鼠心肌受损与水通道蛋白-1(AQP-1)表达状况影响。方法:选取SPF级Wistar雄性大鼠60只,随机数字表法分成四组,正常组、模型组、西药组和观察组,每组各15只,除正常组外其它各组制备癌性腹水大鼠模型;造模以后第四天开始灌胃,西药组灌胃呋塞米药液,剂量为1ml/100g,观察组大鼠灌胃甘遂半夏汤和呋塞米药液,剂量为1ml/100g,模型组、正常组灌胃同剂量蒸馏水,各组大鼠均连续灌胃10天。记录大鼠体重、腹围/体重、尿量及腹水量,ELISA法检测血清醛固酮、钠离子、钾离子、Ure、血清肌酐(Cre)、尿酸(UA)、肌酸激酶(CK)、乳酸脱氢酶(LDH)、丙二醛(MDA)及超氧化物歧化酶(SOD)含量,同时检测大鼠腹膜组织AQP-1mRNA表达状况。结果:模型组、西药组及观察组大鼠体重较正常组下降,腹围/体重较正常上升,差异均有统计学意义(P<0.05);模型组、西药组大鼠醛固酮、钾离子及钠离子含量比正常组上升,AQP-1mRNA表达量比正常组下降,观察组醛固酮、钾离子及钠离子含量比模型组下降,AQP-1mRNA表达量比模型组上升,差异均有统计学意义(P<0.05);模型组、西药组及观察组大鼠Cre含量比正常组上升,Ck含量比正常组下降,模型组与西药组大鼠Ure、UA、LDH比正常组上升,观察组比模型组下降差异均有统计学意义(P<0.05),各组大鼠MDA、SOD差异均无统计学意义(P>0.05)。结论:甘遂半夏汤结合呋塞米可提升癌性腹水大鼠腹膜组织内AQP-1表达量,改善心肌受损状况。
        Objective:To analyze the effect of Gansui Banxia Decoction on the expression of myocardial damage and aquaporin-1(AQP-1) for rats with cancerous ascites.Methods:Sixty Wistar male rats of SPF grade were randomly divided into four groups:normal group,model group,western medicine group and observation group,with 15 rats in each group.The other groups except the normal group prepared cancerous ascites.The fourth day after modeling,the stomach was administered,and the western medicine group was given the furosemide solution at a dose of 1 ml/100 g.The rats in the observation group were intragastrically administered with Ganzi Banxia Decoction and furosemide solution at a dose of 1 ml/100 g,the model group and the normal group were given the same amount of distilled water,and the rats in each group were continuously intragastrically administered for 10 days.Rat body weight,abdominal circumference/body weight,urine volume and ascites volume were recorded.Serum aldosterone,sodium ion,potassium ion,Ure,serum creatinine(Cre),uric acid(UA),creatine kinase(Ck),lactate were detected by ELISA.The contents of hydrogenase(LDH),malondialdehyde(MDA) and superoxide dismutase(SOD) were detected simultaneously,and the expression of AQP-1 mRNA in rat peritoneal tissues was detected.Results:The weight of the rats in the model group,the western medicine group and the observation group decreased compared with the normal group,and the abdominal circumference/body weight increased compared with normal(P<0.05).The aldosterone and potassium ions in the model group and western medicine group.The content of sodium ion was higher than that of the normal group.The expression of AQP-1 mRNA was lower than that of the normal group.The levels of aldosterone,potassium and sodium in the observation group were lower than those in the model group,and the expression of AQP-1 mRNA was higher than that in the model group.The difference was statistically significant(P<0.05);The content of Cre in the model group,western medicine group and observation group increased compared with the normal group,and the Ck content decreased compared with the normal group.The Ure,UA and LDH of the model group and the western medicine group were higher than the normal group,and the observation group was compared with the model group.The difference was statistically significant(P<0.05).There was no significant difference in MDA and SOD between the groups(P>0.05).Conclusion:Ganzi Banxia Decoction combined with furosemide can increase the expression of AQP-1 in peritoneal tissues of rats with cancerous ascites and improve myocardial damage.
