摘要
目的:探讨白藜芦醇对老年雄性大鼠术后认知功能的影响。方法:健康雄性老年SD大鼠36只,采用随机数字表法随机均分成对照组、麻醉手术组和白藜芦醇+麻醉手术组,白藜芦醇+麻醉手术组麻醉手术前连续7 d白藜芦醇100 mg/kg灌胃,另两组给等容量的1%羟甲基纤维素钠溶液灌胃;术后24 h,各组随机取6只大鼠行Morris水迷宫实验测试认知功能,定向航行试验测逃避潜伏期,空间探索实验测目标象限活动时间,另6只大鼠水合氯醛麻醉后断头、冰上迅速剥离海马组织,检测海马组织活性氧(ROS)、丙二醛(MDA)、超氧化物歧化酶(SOD)、肿瘤坏死因子-α(TNF-a)及白细胞介素1β(IL-1β)含量。结果:与对照组比较,麻醉手术组的ROS、MDA、TNF-a、IL-1β含量明显升高,SOD含量明显下降,大鼠术后Morris水迷宫实验结果显示逃避潜伏期明显延长,目标象限活动时间明显缩短,差异有统计学意义(P<0.05);与麻醉手术组比较,白藜芦醇+麻醉手术组的ROS、MDA、TNF-a、IL-1β含量明显下降,SOD含量明显升高;大鼠术后Morris水迷宫实验结果显示逃避潜伏期缩短,目标象限活动时间明显延长,差异有统计学意义(P<0.05)。结论:白藜芦醇可明显降低老年大鼠术后海马组织的氧化应激和炎症反应水平,改善老年大鼠术后认知功能障碍。
        Objective: To investigate the effects of resveratrol on postoperative cognitive dysfunction(POCD) in aged rats. Methods: 36 healthy male SD aged rats were randomly divided into 3 groups(n=12): control group, anesthesia surgerg group and resreratrol anesthesia surgerg group. sevoflurane anesthesia group was given a gavage of resveratrol(100 mg/kg) for 7 consecutive days before surgery, other groups were given an equal volume of 1% sodium carboxymethylcellulose solution. 24 hours after operation, 6 rats were randomly selected from each group. The Morris water maze test was adapted to test the cognitive function, navigation test was chosen to test escape latency, space exploration test was chosen to test quadrant activity time. The other 6 rats were decapitated after chloral hydrate anesthesia. The hippocampus was quickly exfoliated on ice to detect ROS, MDA, SOD, TNF-a and IL-1β content. Results: Compared with control group, the contents of ROS, MDA, TNF-a and IL-1β in anesthesia surgery group were significantly increased, SOD content was significantly decreased, the Morris escape period was prolonged, quadrant activity time was significantly reduced, difference was statistically significant(P<0.05). Compared with anesthesia surgerg group, ROS, MDA, TNF-a and IL-1β content in resveratrol anesthesia group were significantly decreased, SOD was significantly increased; the cruising period of the maze test was shortened, and quadrant activity time was obviously prolonged, difference was statistically significant(P<0.05). Conclusion: Resveratrol can significantly reduce the oxidative stress and inflammatory response in hippocampus of aged rats after surgery and improve postoperative cognitive dysfunction in aged rats.
