摘要
以滑子菇水提醇沉多糖(PNP)为对象,通过傅里叶红外光谱(FT-IR)、核磁共振(1H NMR)、刚果红试验等试验对PNP结构进行分析。体外试验测定了PNP对脾淋巴细胞的增殖和H2O2损伤保护的作用。结构分析表明,PNP具有多糖的特征吸收峰,由4种单糖组成,以β型糖苷键为主不含乙酰基、糖醛酸的吡喃环结构,具有较长的侧链和较多的分枝结构。质量浓度为400μg/mL的PNP对淋巴细胞的增殖作用最大,增殖率为5.93%,100μg/mL及以下浓度时则表现抑制作用。质量浓度为20μg/mL的PNP对H2O2损伤的保护作用最大,保护率为54.66%。
        The polysaccharide of PNP was isolated from Pholiota nameko by ethanol subsiding method.The PNP research on structure were analyzed by FT-IR,1 HNMR,Congo red,et al,and the PNP on Spleen lymphocyte proliferation and H_2O_2 damage of the cells in vitro immune activity were measured.The result showed that PNP had the typical characteristics of polysaccharides absorption peaks,and PNP was composed of four monosaccharides,and the sugar chain was mainly β-configuration pyranoside,no acetyl and uronic acid,and contained a long side chain and branches.PNP had the effect on Spleen lymphocyte proliferation,the best effect on lymphocyte proliferation with 400 μg/mL.The proliferation activity of PNP reached 5.93%,low concentration had no effect on lymphocyte proliferation,the best effect on protection cells of H_2O_2 damage with 20 μg/mL,reached the best protection activity 54.66%.
