葛根素联合乌圆补血口服液对阿尔茨海默病大鼠模型的治疗效果

摘要
目的通过观察葛根素联合乌圆补血口服液治疗阿尔茨海默病大鼠模型的效果,为中西医防治AD提供实验依据。方法取80只6月龄SD大鼠随机分成4组,对照组、模型组、葛根素治疗组和葛根素+乌圆补血口服液治疗组。采用D-半乳糖联合亚硝酸钠造模60 d,再用葛根素和葛根素+乌圆补血口服液对造模后其中两组进行治疗8周。实验结束,用水迷宫实验测大鼠学习记忆力,麻醉大鼠取左心血5 ml,再处死大鼠取大脑。测血清和大脑匀浆谷胱甘肽过氧化物酶(GSH-Px)、超氧化物岐化酶(SOD)的活力和超氧阴离子自由基(O_2~-·)的浓度;测大脑匀浆胆碱脂酶(TchE)活力和丙二醛(MDA)的浓度。结果与模型组比较,两个治疗组血清和大脑匀浆GSH-Px的活力(1.786±0.510,1.768±0.415)、SOD的活力(110.3±10.2,5.892±0.450)明显升高,O_2~-·浓度(6.576±0.605,6.102±0.790)明显降低;而大脑匀浆TchE活力(1.397±0.148)和MDA的浓度(4.894±0.820)明显降低,记忆力明显改善(4.05%);葛根素治疗组和葛根素+乌圆补血口服液治疗组的大脑皮质病理改变得到很好改善,颗粒细胞数量增多,神经元胞质尼氏体也增多。结论较小剂量葛根素联合乌圆补血口服液对AD大鼠模型有治疗效果,且与较大剂量葛根素疗效相近。
        Objective By observing the joint Wuyuan blood puerarin oral treatment of Alzheimer's disease rat model results, A D for the prevention and treatment of Chinese and Western medicine to provide experimental basis. Methods Take 80 six-month-old SD rats were randomly divided into four groups, the control group, model group and treatment group and puerarin puerarin + Wuyuan blood oral treatment groups of 20, Male and female the same number. Joint use of D-galactose nitrite modeling 60 d, and then puerarin and puerarin + Wuyuan blood oral liquid on which the two groups after modeling for 8 weeks. End of the experiment, water maze learning and memory in rats with experimentally measured, take the left narcotic efforts 5 ml, and then were sacrificed brain. Measured in serum and brain homogenates glutathione peroxidase(GSH-PX), superoxide dismutase(SOD) activity and superoxide anion radicals(O_2~-·) of concentrations; Measuring brain homogenate cholinesterase(TchE) activity and malondialdehyde(MDA) concentrations. Results Compared with model group, Two treatment groups in serum and brain homogenates of GSH-PX activity(1.786 ± 0.510,1.768 ± 0.415), SOD vitality(110.3 ± 10.2,5.892 ± 0.450) were significantly increased, O_2~-·Concentrations(6.576 ± 0.605,6.102 ± 0.790) was significantly lower; while the brain homogenates TchE activity(1.397 ± 0.148) and MDA concentrations(4.894 ± 0.820) was significantly lower, Memory was significantly improved(4.05%); Puerarin treatment group and puerarin + Wuyuan blood oral treatment of pathological changes in the cerebral cortex group is well improved, increasing the number of granule cells, neuronal cytoplasmic Nissl bodies also increased. Conclusion Puerarin smaller doses Wuyuan blood oral rat model of AD have a therapeutic effect, and with the larger dose puerarin similar efficacy.
