摘要
目的探讨子宫肌瘤患者孕激素受体(PR)的表达及意义。方法选取2017年1月至2017年12月我院保存的子宫肌瘤组织标本21例,同时选取正常子宫平滑肌组织作为对照,采用免疫组化染色检测PR表达;原代培养子宫肌瘤细胞和正常子宫平滑肌细胞,采用Western-blot检测PR-A、PR-B和PR-M表达,同时筛选出仅表达PR-M的细胞株(PR-M阳性组)和仅表达PR-A/B的细胞株(PR-M阴性组)进行CCK-8实验。结果子宫肌瘤组织PR阳性表达率为95.24%,明显高于正常子宫平滑肌组织(P<0.05);子宫肌瘤细胞PR-A、PR-B、PR-M蛋白相对表达量分别为(0.762±0.116)、(0.987±0.107)和(0.097±0.042),明显高于子宫平滑肌细胞(P<0.05);PR-M阳性组培养24h、48h吸光值(OD)分别为(0.352±0.097)和(0.411±0.091),明显高于PR-M阴性组(P<0.05)。结论子宫肌瘤组织以及细胞PR表达升高,其中PR-M蛋白可能与子宫肌瘤细胞增殖有关,值得进一步研究。
        Objective To investigate the expression and significance of progesterone receptor(PR) in patients with uterine fibroids. Methods A total of 21 specimens of hysteromyoma preserved in our hospital from January 2017 to December 2017 were selected and normal uterine smooth muscle tissue was selected as the control,the expression of PR was detected with immunohistochemical staining;The myoma cells and normal uterine smooth muscle cells were primary cultured,the expression of PR-A,PR-B and PR-M were detected by using Western-blot,the cell line(PR-M positive group) only expressing PR-M and the cell line only expressing PR-A/B(PR-M negative group) were screened out for CCK-8 test. Results The positive expression rate of PR in hysteromyoma was 95.24%,which was significantly higher than that of normal uterine smooth muscle tissue,and the difference was statistically significant(P<0.05);The relative expressions of PR-A,PR-B and PR-M in hysteromyoma cells were(0.762±0.116),(0.987±0.107) and(0.097±0.042),which were significantly higher than those in uterine smooth muscle cells(P<0.05);The absorbance values(OD) at 24 h and 48 h in PR-M positive group were(0.352±0.097) and(0.411± 0.091),which were significantly higher than those in PR-M negative group,and the difference was statistically significant(P<0.05). Conclusion The expression of PR in hysteromyoma tissues and cells are increased,and PR-M protein may be related to the proliferation of myoma cells,is worth further study.
