摘要
目的:观察逍遥散对肝纤维化模型大鼠的肝功能、肝纤维化指标的影响。方法:选取清洁级成年Wistar大鼠80只,采用随机数字表法分为空白组(等量生理盐水)、模型组(等量生理盐水)、阳性对照组(秋水仙碱)、逍遥散低剂量组(1 g·mL~(-1)的逍遥散溶液,按照8 g·kg~(-1)灌胃)、逍遥散高剂量组(1 g·mL~(-1)的逍遥散溶液,按照16 g·kg~(-1)灌胃),均连续给药8周。结果:干预8周后,逍遥散高剂量组大鼠血清丙氨酸氨基转移酶(alanine aminotransferase,ALT)、天门冬氨酸氨基转移酶(aspartate aminotransferase,AST)水平均低于阳性对照组和逍遥散低剂量组(P<0.05),白蛋白(albumin,ALB)高于阳性对照组和逍遥散低剂量组(P<0.05)。逍遥散高剂量组的血清透明质酸(hyaluronic acid,HA)、层黏蛋白(laminin,LN)、Ⅲ型前胶原(precollagen typeⅢ,PCⅢ)、Ⅳ型胶原(collagen typeⅣ,Ⅳ-C)水平低于阳性对照组和逍遥散低剂量组(P<0.05)。逍遥散高剂量组的转化生长因子β1(transforming growth factor-β1,TGF-β1)、血小板衍生生长因子(platelet-derived growth factor,PDGF)蛋白表达水平低于阳性对照组和逍遥散低剂量组(P<0.05)。结论:逍遥散能显著改善肝纤维化模型大鼠肝功能(降低ALT、AST水平)和血清肝纤维化指标(降低HA、LN、PCⅢ及Ⅳ-C水平),降低肝组织中TGF-β1及PDGF蛋白表达水平。
        Objective:To observe the effects of Xiaoyao Powder on liver function and liver fibrosis in rats with liver fibrosis.Methods:Eighty adult Wistar rats were randomly divided into a blank group(equal saline),a model group(equal saline),a positive control group(colchicine treatment),and a low volume.The Xiaoyao Powder low dose group(1 g·mL~(-1) Xiaoyao Powder) solution gavage was carried out with 8 g·kg~(-1),while Xiaoyao Powder high dose group(1 g·mL~(-1) Xiaoyao Powder) solution gavage was carried with 16 g·kg~(-1).The drug was administered continuously for 8 weeks.RESULTS:After 8 weeks of intervention,the serum levels of alanine aminotransferase(ALT) and aspartate aminotransferase(AST) in the high dose group of Xiaoyao Powder were lower than those of the positive control group and Xiaoyao Powder low dose group(P<0.05),while albumin(ALB) was higher than the positive control group and Xiaoyao Powder low dose group(P<0.05).Serum hyaluronic acid(HA),laminin(LN),type Ⅲ procollagen(precollagen type Ⅲ,PCⅢ),type Ⅳ collagen(collagen type Ⅳ,Ⅳ-C) levels in high-dose group.It was lower than the positive control group and Xiaoyao Powder low dose group(P<0.05).The expression levels of transforming growth factor-β1(TGF-β1) and platelet-derived growth factor(PDGF) in the high-dose group were lower than those in the positive control group and Xiaoyao Powder low-dose group(P<0.05).Conclusion:Xiaoyao Powder can significantly improve liver function(reduced ALT,AST level) and serum fibrosis index(reduced HA,LN,PCⅢ and Ⅳ-C levels) in liver fibrosis model rats,and reduce TGF-β1 in liver tissue as well as PDGF protein expression level.
