摘要
目的:制备可明显吸附卵清蛋白(OVA)的Al(OH)_3佐剂并用其建立具备变应性鼻炎(AR)临床客观特征的大鼠模型.方法:通过控制反应条件制备可明显吸附OVA的Al(OH)_3佐剂,对模型组用OVA辅以自制Al(OH)_3佐剂进行基础致敏,继以质量分数为2%OVA生理盐水行鼻内激发.对照组以生理盐水替代自制Al(OH)_3佐剂.采用PCA、行为学评分、Luna染色、HE染色评价模型.结果:1)吸附研究表明自制Al(OH)_3佐剂对OVA具明显吸附作用;2)模型组:PCA(+)证明变应原特异性Ig E产生;行为学评分>5分;Luna染色与HE染色示鼻黏膜嗜酸性粒细胞增多、浸润.对照组无上述变化.结论:所制Al(OH)_3佐剂对OVA具明显吸附作用并在此基础上成功建立了具备Ig E阳性和鼻黏膜嗜酸性粒细胞增多AR临床客观特征的大鼠模型.
        Aim: Prepared a new kind of Al(OH)_3adjuvant which has obviously absorbability on ovalbumin,and applied in establishing a rat model with objective clinical characteristics of allergic rhinitis(AR). Methods: Firstly, a new kind of Al(OH)_3adjuvant with obvious adsorption on ovalbumin(OVA) had been prepared by controlling the reaction conditions,then,was used in building AR rat model. The model group was treated by ovalbumin and Al(OH)_3to complete basic sensitization;followed by nasal challenge with 2% ovalbumin saline. The control group used normal saline instead of Al(OH)_3adjuvant. By the passive cutaneous anaphylaxis, the behavioral score, Luna staining,Hematoxylin-eosin staining,the model was comprehensively evaluated. Results: 1) the results of OVA absorption experiments showed our Al(OH)_3could absorb OVA obviously. 2) the model group: passivecutaneous anaphylaxis(+) proved the production of allergen specific Ig E; behavioral score was greater than 5; Luna staining and Hematoxylin-eosin staining showed the nasal mucosa eosinophilia infiltration.The control group had no such changes. Conclusion: Based on the preparation of Al(OH)_3adjuvant with obvious adsorption on ovalbumin,we had successfully established the rat model with allergen specific Ig E(+) and nasal mucosa eosinophilia infiltration two objective clinical characteristics of allergic rhinitis.
