摘要
目的应用多参数流式细胞术检测急性白血病细胞表面CD抗原分子的表达水平,探讨AML和ALL中CD抗原的表达规律,为临床个体化治疗累积分子机制研究材料。方法应用流式细胞术检测44例急性髓细胞白血病患者和18例急性淋巴细胞白血病患者,以CD45/SSC设门,应用13种单克隆抗体对CD45弱表达的细胞群进行免疫分型,测定多种CD分子的表达率,并通过χ~2检验比较不同的高表达CD分子的相关性。结果 44例AML患者中表达且高表达CD33(79.6%),CD38(70.5%),CD117(52.3%),cyt-MPO(50.0%);16例B-ALL患者中CD10(75.0%),CD19(81.25%)以及CD22(81.25%)阳性表达率较高;比较AML患者高表达CD抗原,结果显示CD33/CD38、CD33/cyt-MPO的抗原表达差异均有统计学意义(P<0.05)。结论不同的CD分子在急性白血病细胞上表达有差异性,或将为靶向治疗和临床个体化治疗提供有用的线索。
        Objective To detect the expression level of CD antigen on the surface of acute leukemia cell by multiparameter flow cytometry,and to explore the expression principle of CD antigens in AML and ALL,so as to accumulate molecular mechanism research materials for clinical individualized treatment.Methods 44 patients with acute myeloid leukemia and 18 patients with acute lymphoblastic leukemia were detected by flow cytometry.Based on CD45/SSC,13 types of monoclonal antibodies were used to carry out immunophenotype of the cell population with weak expression of CD45,and expression rate of various CD molecules was determined.The χ~2 test was used to compare the correlation of different highly expressed CD molecules.Results CD33(79.6%),CD38(70.5%),CD117(52.3%) and cyt-MPO(50.0%)were highly expressed in 44 patients with AML;the positive rate of CD10(75.0%),CD19(81.25%)and CD22(81.25%)was higher in 16 patients with B-ALL;the high expression of CD antigens in the patients with AML were compared.The results showed that there was a statistically significant difference in the antigen expression of CD33/CD38 and CD33/cyt-MPO(P<0.05).Conclusion Different CD molecules are differentially expressed on acute leukemia cells,or they will provide useful clues for targeted therapy and clinical individualized treatment.
