摘要
目的:研究槲皮素与芦丁的薄层色谱-表面增强拉曼光谱(TLC-SERS),用于中药中有效成分的鉴别。方法:通过实验考察,确定了激发光波长;采用拉曼微测技术分别扫描涂有表面增强剂的槲皮素与芦丁TLC斑点,获得二维平面分布像图,将激光束快速定位于样品与表面增强剂活位结合点上,得到相应的SERS;并与常规拉曼光谱对比。结果:以780 nm激光为激发波长,银溶胶为表面增强剂,通过比较分析槲皮素与芦丁的TLC-SERS光谱,建立了黄蜀葵花、槐花中有效成分的二维TLC-SERS鉴别法。结论:本方法专属性强,灵敏度高,重复性好,可用于中药的鉴别。
        Objective:To study the TLC-SERS of quercetin and rutin for the identification of active ingredients in traditional Chinese medicine. Methods:Through the experimental investigation,the wavelength of the excitation light was selected.The Raman micro-measurement technique was used to scan the spots of quercetin and rutin coated with surface enhancer toobtaina two-dimensional distribution image plane. The corresponding conding point of SERS which was produced by rapid positioning of the laser beam on the sample surface with the enhancer-binding activities was obtained.And then,the results were compared with the ones acquired by regular Raman spectroscopy. Results:The TLC-SERS of quercetin and rutin were established with excitation wavelength at 780 nm,surface enhancer of silver colloids and micro-Raman measurement technology,further successfully applied to identify quercetin in Abelmoschi Corolla and rutin in Sophorae Flos.Conclusion:The method established in this study is found to have significant specificity,high sensitivity and good reproducibility,and can be applied to the identification of traditional Chinese medicine.
