芍药苷对脂多糖诱导的人牙周膜细胞损伤的保护作用及机制研究

摘要
目的:探讨芍药苷对脂多糖(LPS)诱导的人牙周膜细胞(hPDLCs)损伤的保护作用及机制。方法:分离hPDLCs并鉴定和培养,将hPDLCs分为对照组、模型组和芍药苷低、中、高剂量组,芍药苷作用24h后测定各组细胞增殖率,检测细胞中白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)、肿瘤坏死因子-ɑ(TNF-ɑ)、基质金属蛋白酶-2(MMP-2)和基质金属蛋白酶-9(MMP-9)表达水平。结果:与对照组比较,模型组和各芍药苷剂量组细胞增殖率降低,hPDLCs细胞中IL-6、IL-1β、TNF-ɑ、MMP-2和MMP-9水平增高,差异均具有统计学意义(P<0.05);与模型组比较,各芍药苷剂量组细胞增殖率增加,且随着芍药苷剂量的增加,增加幅度越大,hPDLCs细胞中IL-6、IL-1β、TNF-ɑ、MMP-2和MMP-9水平降低,且随着芍药苷剂量的增加,增加/降低幅度越大,差异均具有统计学意义(P<0.05)。结论:芍药苷能抑制炎症因子(IL-6、IL-1β、TNF-ɑ)和基质金属蛋白酶-2、9的表达,减轻LPS诱导的hPDLCs细胞损伤。
        Objective: To investigate the protective effect and mechanism of paeoniflorin in lipopolysaccharide( LPS)-induced injury of human periodontal ligament cells( hPDLCs). Methods: After the identification and culture,hPDLCs were divided into five groups including the control group,the model group,and low-dose,middle-dose and high-dose paeoniflorin groups. The cell proliferation rate and the expression levels of interleukin( IL-6),interleukin-1β( IL-1β),tumor necrosis factor-α( TNF-α),matrix metalloproteinase-2( MMP-2) and matrix metalloproteinase-9( MMP-9) were detected and compared among the groups. Results: Compared with that in the control group,the cell proliferation rate of the model group and each paeoniflorin group decreased,while the levels of IL-6,IL-1 beta,TNF-α,MMP-2 and MMP-9 in hPDLCs cells increased with statistical significance( P < 0. 05). Compared with that in the model group,the cell proliferation rate of each paeoniflorin group increased,and was positively correlated with the dose of paeoniflorin. With the increase of paeoniflorin dosage,the levels of IL-6,IL-1 beta,TNF-α,MMP-2 and MMP-9 in hPDLCs decreased. The increase/decrease was greater when the dosage of paeoniflorin was increased,and the difference was statistically significant( P <0. 05). Conclusion: The protective effect and mechanism of paeoniflorin in LPS-induced injury of hPDLCs may be related to the decrease of IL-6,IL-1β,TNF-α,MMP-2 and MMP-9.
