淫羊藿苷和骨髓间充质干细胞共育液对兔骨关节炎模型关节液中IL-1、TNF-α、IL-10、MMP-3表达的影响

摘要
目的观察淫羊藿苷与骨髓间充质干细胞共育液对兔骨关节炎模型关节液中IL-1、TNF-α、IL-10、MMP-3表达的影响。方法将20只兔随机分为空白组、模型组、BMSCs组、BMSCs+ICA组。造模成功后BMSCs组和BMSCs+ICA组关节腔内分别注射BMSCs和BMSCs+ICA;空白组和模型组注射等量生理盐水。结果与对照组相比,各组治疗前关节液中IL-1、TNF-α、MMP-3水平升高,IL-10水平下降。与模型组相比,BMSCs及BMSCs+ICA组关节液中IL-1、TNF-α、MMP-3水平下降,IL-10水平升高,且BMSCs+ICA组改善幅度优于BMSCs组。结论淫羊藿苷和骨髓间充质干细胞共育液关节腔注射能降低兔骨关节炎模型关节液中IL-1、TNF-α、MMP-3的表达,升高IL-10的表达。
        Objective To observe the effect of intraarticular injection of icariin and bone marrow mesen-chymal stem cells on the expression of IL-1,TNF-α,IL-10 and MMP-3 in the articular fluid of rabbit osteoarthritismodel. Methods Twenty rabbits were randomly divided into four groups. After successful modeling,the BMSCsgroup was injected with BMSCs,BMSCs + ICA into the articular cavity,and the blank group was injected with thesame amount of normal saline. Results Compared with the control group,IL-1,TNF-α,MMP-3 levels were in-creased and IL-10 levels were decreased in the other groups before the treatment. Compared with the model group,IL-1,TNF-α,MMP-3 were decreased and IL-10 level increased in BMSCs group and BMSCs + ICA group. BMSCs+ ICA group was better than BMSCs groups. Conclusion The results show that icariin and bone marrow mesenchy-mal stem cells are incubated in liquid intra-articular injection can reduce the rabbit osteoarthritis IL-1,TNF-α,MMP-3 expression and increased IL-10 expression.
