miR-143与骨肉瘤的研究进展

摘要
随着新辅助化疗结合保肢术方法的完善,骨肉瘤患者5年生存率有所提高,但是化疗抵抗和肺转移仍是骨肉瘤不良预后的重要原因和难以逾越的屏障。近年来,随着微RNA(miRNA)与骨肉瘤发病机制关系的研究逐年增加,发现骨肉瘤患者中miRNA表达的差异性与其发病密切相关;同时发现miRNA对骨肉瘤的诊断和预后的判断有一定的意义。其中,miRNA-143(miR-143)可能作为抑癌基因参与了骨肉瘤的发生、发展。骨肉瘤发病与体内miR-143下降密切相关,且伴随着miR-143基因表达升高,骨肉瘤细胞的增殖、凋亡以及侵袭和转移的能力有所改变。本文阐述了具有预后标志物潜能的miR-143调控的基因(K-ras、Bcl-2、PAI-1等)与骨肉瘤发生、发展和转移之间的关系。
