头穴丛刺结合跑台训练对坐骨神经损伤大鼠小腿三头肌萎缩调控及Murf 1蛋白表达影响

摘要
目的:探讨头穴丛刺结合跑台训练对坐骨神经损伤大鼠神经功能恢复、小腿三头肌萎缩,以及肌肉环状指蛋白1(Murf 1)蛋白表达的影响。方法:清洁级大鼠84只,随机分为5组,其中空白组、假手术组分别6只,模型组、跑台组和头穴丛刺结合跑台组,每组24只,并且每组分为7d、14d、21d、28d四个亚组。采用钳夹法造模,假手术组要充分暴露坐骨神经但不钳夹。造模3天后进行干预,头穴丛刺结合跑台组:头穴丛刺+跑台训练,跑台组:跑台训练,其余3组除定时抓取外不做任何干预。各组相应时间点需要测定坐骨神经功能指数(SFI);小腿三头肌湿重、Western Blotting检测泛素蛋白连接酶Murf 1蛋白的表达。结果:(1)小腿三头肌湿重比和坐骨神经功能指数均下降,与模型组比较,头穴丛刺结合跑台组和跑台组均高于模型组(P<0.05);且头穴丛刺结合跑台组高于跑台组(P<0.05);(2)Murf 1蛋白的水平明显增加,与模型组比较,跑台组和头穴丛刺结合跑台组均低于模型组(P<0.05),且头穴丛刺结合跑台组低于跑台组(P<0.05)。结论:头穴丛刺结合跑台训练以及跑台训练均能够提高SFI,下调Murf 1蛋白水平,减少小腿三头肌湿重比下降,延缓肌肉萎缩,提高坐骨神经功能,并且头穴丛刺结合跑台训练优于单纯跑台训练。
        Objective: To explore the effects of cluster needling of scalp point combined with treadmill training on neural functional recovery, the atrophy of triceps surae and the expression of muscle ring finger protein 1(Murf 1)protein in rats with sciatic nerve injury.Method: Eighty-four rats of clean grade were randomly divided into 5 groups, including the model group, the treadmill group and the cluster needling of scalp point combined with treadmill group, 24 rats in each group,and each group was divided into 7 d, 14 d and 21 d, 28 d subgroups. We also established the blank group and the control group with 6 rats in each group. The clamps method was used to build model and fully exposed sciatic nerve with no clamps was performed the in control group.After 3 d The intervention was carried out in3 days after model was made: the cluster needling of scalp point combined with treadmill group: the cluster needling of scalp point+ treadmill training, the treadmill group: treadmill training, the other three groups :no intervention except timing grab.The sciatic nerve function index(SFI), the wet weight of triceps surae and the expression of Murf 1 protein were detected at each corresponding time points.Result:(1)The wet weight ratio of triceps surae and the sciatic nerve function index decreased and was higherin the treadmill group and the cluster needling of scalp point combined with treadmill group when compared with the model group(P<0.05). Furthermore the ratio and index were higher in the cluster needling of scalp point combined with treadmill group than that in the treadmill group(P<0.05);(2)The Murf 1 protein level increased significantly. The protein level was lower in the treadmill group and the cluster needling of scalp point combined with treadmill group compared with model group(P<0.05). And the cluster needling of scalp point combined with treadmill group also had the lower Murf 1 protein level than that in the treadmill group(P<0.05).Conclusion:Both the treadmill training and the cluster needling of scalp point combined with treadmill training can improve the SFI, down-regulate Murf 1 protein level, prevent the decrease of wet weight ratio in triceps surae, delay muscle atrophy and enhance the sciatic nerve function. Especially, the cluster needling of scalp point combined with treadmill training has better effect that the treadmill training only.
