摘要
目的探讨蛇黄肝炎汤含药血清对人肝癌细胞HepG-2增殖的影响。方法 25只大鼠随机分为1倍、2倍、4倍浓度含药血清组,复方环磷酰胺含药血清组(阳性药物血清组),空白血清组5个实验药物组,分别将不同浓度的蛇黄肝炎汤含药血清、阳性药物含药血清、空白血清与HepG-2细胞共同培养,采用四甲基偶氮唑蓝(MTT)比色法及细胞集落形成实验观察蛇黄肝炎汤含药血清体外对HepG-2细胞增殖、克隆形成能力的影响。结果与空白血清组相比较,2、4倍的蛇黄肝炎汤含药血清组吸光度值(OD值)均降低,差异均有统计学意义(P<0.05)。4倍含药血清随作用时间的延长,对HepG-2细胞的抑制率逐渐增加(P<0.05),当4倍浓度含药血清抑制时间为72 h时,抑制率达76.82%,与阳性药物血清组作用相当(抑制率达77.71%)。不同浓度的含药血清均能有效降低肿瘤细胞集落形成,与空白血清组相比差异均有统计学意义(P<0.05),且随着含药血清浓度增大细胞集落形成数逐渐降低,集落形成抑制率随含药血清浓度的增加呈逐渐升高的趋势。结论蛇黄肝炎汤含药血清具有抑制人肝癌细胞HepG-2体外增殖、降低其克隆形成能力作用。
        Objective To explore the impacts of shehuang ganyan decoction- contained serum on human Hepg- 2 proliferation. Methods Twenty- five rats were randomized in the drug- contained serum groups of 1- time,2- time and 4- time concentrations,a compound cytoxan- contained serum group( the positive drug serum group and a blank serum group. HepG- 2 cells were cultivated with corresponding serum in each group. MTT colorimetry and cell colony formation experiment were used to observe the impacts of shehuang ganyan decoction- contained serum on in vitro HepG- 2 proliferation and clonality. Results Compared with the blank serum group,OD value was reduced in the shehuang ganyan decoction- contained serum groups of 2- time and 4- time concentrations,indicating the significant difference( P < 0. 05). With the effect duration of 4- time drug- contained serum,the inhibitive rate of HepG- 2 cells was increased gradually( P < 0. 05). In 72 h,the inhibitive rate was up to 76. 82%,similar to 77. 71% in the positive drug serum group. The drug- contained serum of different concentrations all reduced effectively the colony formation of cancer cell,presenting the significant differences as compared with the blank serum group( P < 0. 05). With the increase of the drug- contained concentration,the number of cell colony formation was reduced gradually and the inhibitive rate of colony formation was increased in tendency with the increase of drug- contained serum concentration. Conclusion The shehuang ganyan decoction- contained serum inhibits in vitro proliferation of HepG- 2 and reduces clonality.
