摘要
目的分析健康对照组和脑梗死患者循环circRNA表达谱的差异性,研究脑梗死的表观遗传学发病机制,为寻找治疗脑梗死的新靶点提供一定的理论依据。方法选取2016年12月~2017年12月入住我院神经内科的急性脑梗死患者40例作为脑梗死组,并选取体检中心健康人群40例作为健康组。脑梗死组于治疗前(脑梗死发生72小时内)于清晨空腹时抽取血标本5 mL,健康组于清晨空腹时抽取血标本5 mL。各血清样本通过高通量技术测定循环circRNA表达谱并分析其差异性,对有显著差异的circRNA建立GO、KEGC分析。结果与健康组相比,有348种已知的差异表达的circRNA,其中有186种上调,162种下调,通过生物信息学分析,脑梗死患者血液中差异表达的circRNA主要富集于mRNA监视通路、胆碱能信号通路等。结论脑梗死组患者的circRNA表达谱与健康组有差异,支持circRNA可能与脑梗死的发生、发展和修复有着密切的关系。
        Objective To analyze the difference of circulating circRNA expression profile between healthy control group and cerebral infarction patients,to study the epigenetic pathogenesis of cerebral infarction,and to provide a theoretical basis for finding new targets for the treatment of cerebral infarction.Methods 40 patients with acute cerebral infarction were selected from December 2016 to December 2017 as cerebral infarction group,and40 healthy people in physical examination center were selected as healthy group.The blood samples of cerebral infarction group were collected at fasting in the morning before treatment(within 72 hours of cerebral infarction),and5 ml at fasting in healthy group in the morning.The circulating circRNA expression profile was determined by high throughput technique and its difference was analyzed. GO,KEGC analysis was established for circRNA with significant difference.Results Compared with the control group,there were 348 known differentially expressed circRNA,of which 186 were up-regulated and 162 were down-regulated.By bioinformatics analysis,the differential expression of circRNA in the blood of cerebral infarction patients was mainly concentrated in mRNA monitoring pathway,cholinergic signal pathway and so on.Conclusion The expression profile of circRNA in cerebral infarction group was different from that in healthy group,indicating that circRNA might be closely related to the occurrence,development and repair of cerebral infarction.
