优质耐阴观叶植物熊掌木的离体快繁研究

摘要
以熊掌木当年生嫩茎为外植体,以MS培养基为基础培养基,附加不同种类和浓度的植物生长调节物质诱导丛生芽及再生植株,建立熊掌木的高效再生繁殖体系。结果表明:先用75%酒精浸泡30 s,再用0.1%HgCl_2浸泡6 min的灭菌效果最佳;最佳增殖培养基为MS+6-BA 4 mg·L~(-1)+NAA 0.2 mg·L~(-1);在培养基MS+6-BA 4 mg·L~(-1)+NAA 0.2 mg·L~(-1)+蔗糖3.0%+琼脂0.7%+AgNO_30.1 mg·L~(-1)上,试管苗玻璃化程度最低,且芽增殖倍数最高;最佳生根培养基为1/2MS+NAA 0.1 mg·L~(-1)+IBA 0.01 mg·L~(-1),试管苗根系质量最好;将试管苗移栽在河沙︰珍珠岩︰草炭土(1︰1︰1)的基质上,成活率高达94%。
        In this study,to establish the high frequency propagation system of Fatshedrea lizei,the annual tender steem of Fatshedrea lizei was taken as explant and MS medium as basic medium,multiple shoots and regeneration plants were induced by adding differernt kinds and concentrations of plant growth regulators.Rusult indicated:the best sterilization procedure for shoot tip was soaked in 75%alcohol for 30s and followed by 0.1%HgCl_26 minutes.The optimum medium for bud proliferation was MS medium with 6-BA4 mg·L~(-1)+NAA0.2 mg·L~(-1),which could produce a large number of adventitious buds.In the MS medium with 6-BA4 mg·L~(-1)+NAA0.2 mg·L~(-1)+Sucrose 3.0%+Agar 0.7%+AgNO_30.1 mg·L~(-1),the vitrification degree of Fatshedrea lizei was the lowest and the bud proliferation was the highest.The optimum rooting medium was half MS medium with NAA 0.1 mg·L~(-1)+IBA 0.01 mg·L~(-1).The transplant survival rate was high on the transplant medium composed of sand:pearlite:peat=1:1:1,survival rate was up to 94%.
