两种多囊卵巢综合征伴胰岛素抵抗大鼠模型的比较研究

摘要
目的分别采用胰岛素(INS)联合绒毛膜促性腺激素(HCG)法、来曲唑(LE)法建立大鼠PCOS动物模型,探讨其生殖和代谢异常的特征并加以比较。方法将实验大鼠随机分为3组:A组(对照组)、B组(INS+HCG组)、C组(LE组)。观察动情周期及体质量变化;造模后取血检测性激素及糖脂代谢指标;卵巢组织切片HE染色观察局部形态学改变。结果 B组和C组体质量和卵巢质量增加(P<0.05);A组有规律动情周期,B组及C组动情周期紊乱;A组卵巢局部形态基本正常,B组和C组均出现卵巢多囊样改变;B组T、LH、LH/FSH、FPG、FINS、HOMA-IR和TG均显著增高(P<0.05),C组T、FINS、HOMA-IR增高(P<0.05);B组FPG、FINS、HOMA-IR均高于C组(P<0.05)。结论 INS+HCG法和LE法均可成功诱导大鼠PCOS胰岛素抵抗模型,INS+HCG法诱导的大鼠模型其生殖及代谢指标的变化较LE更具优势。
        Objective Animal models of polycystic ovarian syndrome(PCOS) in rats were established by insulin(INS)+chorionic gonadotropin(HCG) method and letrozole(LE) gavage method respectively, and to discuss and compare the characteristics of abnormal reproduction and metabolism. Methods The experimental rats were randomly divided into 3 groups: group A(control), group B(INS+HCG), group C(LE). The changes of estrous cycle and body mass were observed. The blood samples were taken to detect the sex hormone and glucose and lipid metabolism parameters. The local morphological changes were observed by HE staining in ovarian tissue sections. Results Body mass and ovarian mass increased in group B and C(P<0.05). Estrous cycle was regular in group A, and that was disordered in group B and C. Local ovarian morphology was normal in group A, and ovarian polycystic changes were found in group B and C. T, LH, LH/FSH, FPG, FINS, HOMA-IR and TG were significantly increased in group B(P<0.05), and T, FINS, HOMA-IR and TG were significantly increased in group C(P<0.05). FPG, FINS and HOMA-IR in group B were higher than those in group C(P<0.05). Conclusion Both INS+HCG and LE method could successfully induce the rat model of PCOS insulin resistance. But the changes of reproductive and metabolic parameters in rat models induced by INS+HCG method were more advantageous than LE method.
