太赫兹波对MG-63荷瘤小鼠快速活体成像的实验研究

摘要
目的观察太赫兹波在人成骨肉瘤细胞株MG-63荷瘤小鼠活体瘤体组织显像中的应用效果,并探讨太赫兹波技术在肿瘤组织切除术中的指导作用。方法利用华中科技大学武汉光电国家实验室连续太赫兹波成像系统,进行人成骨肉瘤细胞株MG-63荷瘤小鼠活体的瘤体显像。实验组小鼠(8只)在太赫兹成像系统指导下进行瘤体切除术,对照组(8只)常规肉眼下行瘤体切除术,术后将切除的肿瘤组织和切除肿瘤后的边界组织送病理切片;继续观察两组小鼠术后的生长情况以及肿瘤原位复发情况。结果人成骨肉瘤MG-63荷瘤小鼠在太赫兹波成像检测下,瘤体组织处出现强烈吸收信号,吸收强度明显高于健康组织;实验组术后原瘤体部位的强吸收信号消失,对照组术后原瘤体部位的吸收信号明显减弱,但周边仍存留散在较强的吸收区域。实验组切除组织边缘取材的肿瘤阳性率为8.3%,远低于对照组的37.5%,差异有统计学意义(P<0.05)。后续观察中,实验组和对照组分别死亡1只、2只小鼠,两组死亡率的差异无统计学意义,但两组的肿瘤复发率的差异存在统计学意义(P=0.038)。结论太赫兹波成像技术能对MG-63荷瘤小鼠中骨肉瘤肿瘤组织进行活体即时显像,能指导有效手术切除恶性肿瘤组织,具有很好的临床应用前景。
        Objective To investigate the terahertz wave of fast imaging situation in MG-63 tumor-bearing mice, and to explore the role of terahertz wave technique in osteosarcoma tumor resection. Methods With the continuous terahertz imaging system which come from Wuhan National Laboratory for Optoelectronicsof Huazhong University of Science and Technology, the continuous terahertz imaging in MG-63 tumor-bearingmice in vivo was performed. Eight mice in the experimental group underwent tumor resection under theguidance of terahertz imaging, and 8 mice in control group were treated with routine naked eye resection. Theresectedtissues were subjected to pathological analysis. The mice were kept for one week and the growth of themice in the two groups was observed and the presence of tumor recurrence was observed. Results The MG-63 tumor-bearing mice showed a strong absorption signal at the tumor tissue under terahertz imaging, which wassignificantly different from that of the normal tissues. The strong absorption signal of the tumor site disappearedin the experimental group after operation,and that in the control group was obviously weakened,but the diffusestrong absorption areastill remained inthe surrounding region.The positive rate of tumor in the margin ofresected tissues was 8.3% in the experimental group, significantly lower than in the control group(8.3% vs.37.5%)(P<0.05). There were one and two deaths in the experimental group and the control group, respectively.There was significant difference in the recurrence rate between the two groups(P=0.038). Conclusion Terahertz wave technique can perform real-time imaging of osteosarcoma tumor tissue in MG-63 tumor-bearingmice in vivo, can effectively guide the treatment of surgical resection of malignant tumor tissue, and has a goodclinical application prospect.
