摘要
目的:分析针刺对急性脑缺血大鼠脑缺血损伤及能量代谢的影响。方法:选取SPF级Wistar雄性大鼠30只,随机数字表法将大鼠分成三组,正常组(10只)、模型组(10只)及观察组(10只),模型组和观察组大鼠依据微创开颅法对大脑动脉电凝,以制备急性缺血大鼠模型,观察组在大鼠造模2小时清醒后行针刺治疗,选取水沟穴、百会穴,两个穴位行捻转泻法,频率为120次/分,每次1分钟,每分钟5分钟进行1次,留针30分钟。模型组和正常组大鼠正常喂养,不进行针刺。高效液相检测大鼠缺血脑组织内一磷酸腺苷(AMP)、二磷酸腺苷(ADP)及三磷酸腺苷(ATP)含量,比色法检测脑组织内脑组织内Ca2+-ATP、Na+-K+-ATP酶含量。结果:模型组和观察组大鼠脑组织内AMP含量较正常组升高,ADP、ATP含量较正常组降低,观察组升高、降低幅度低于模型组,差异均有统计学意义(P<0. 05);模型组大鼠脑组织内Ca2+-ATP、Na+-K+-ATP酶含量较正常组降低,观察组大鼠脑组织内Ca2+-ATP、Na+-K+-ATP酶含量较模型组升高,差异均有统计学意义(P<0. 05)。结论:急性脑缺血大鼠行针刺治疗可明显降低其脑缺血损伤,改善脑组织能量代谢障碍。
        Objective: To analyze the effect of acupuncture on cerebral ischemic injury and energy metabolism in rats with acute cerebral ischemia. Methods: Thirty Wistar male rats were selected and randomly divided into three groups: normal group( 10),model group( 10),and observation group( 10). The model group and the observation group were large. Rats were subjected to electrocoagulation in the cerebral arteries according to the minimally invasive craniotomy to prepare an acute ischemic rat model. The observation group was acupunctured after the rats had been awake for 2 hours after the model was established. The Shuigou and Baihui points were selected and the two points were selected. The procedure was performed at a rate of 120 beats per minute,one minute at a time,five times a minute,and a needle for 30 minutes. The rats in the model group and the normal group were fed normally without acupuncture. The contents of adenosine monophosphate( AMP),adenosine diphosphate( ADP)and adenosine triphosphate( ATP) in the ischemic brain tissue of rats were detected by HPLC. The colorimetric method was used to detect intracellular Ca2+-ATP,Na+-K+-ATPase in brain tissue. Results: Compared with the normal group,the AMP content in the brain tissue of the model group and the observation group was higher than that of the normal group. The ADP and ATP contents were lower than the normal group,and the increase and decrease in the observation group was lower than that in the model group. The difference was statistically significant( P < 0. 05). The contents of Ca2+-ATP and Na+-K+-ATPase in the brain tissue of the model group were lower than those in the normal group. The contents of Ca2+-ATP and Na+-K+-ATPase in the brain of the rats in the observation group were higher than those in the model group. The differences were statistically significant( P < 0. 05). Conclusion: Acupuncture treatment in rats with acute cerebral ischemia can significantly reduce cerebral ischemic injury and improve brain energy metabolism.
