摘要
分别采用稀碱-酶解法和稀碱-浓盐法提取鹿气管中的硫酸软骨素,通过比较提取率、提取时间等对2种方法进行考察,最终选取提取率高、耗时短的稀碱-浓盐法提取其中的硫酸软骨素。采用高效液相色谱法对鹿气管中硫酸软骨素含量进行测定,并探讨鹿气管硫酸软骨素及硫酸软骨素寡糖的抗氧化活性。结果发现,稀碱-浓盐法提取鹿气管中硫酸软骨素的得率为21.88%;鹿气管中总硫酸软骨素含量为35.31 g/kg,硫酸软骨素A、B和C含量分别为19.54、2.06 g/kg和13.71 g/kg;鹿气管硫酸软骨素及其寡糖均具有一定的抗氧化活性,且硫酸软骨素寡糖的抗氧化活性显著高于硫酸软骨素。
        Dilute alkali-enzymatic hydrolysis method and dilute alkali-concentrated salt method were used to extract chondroitin sulfate from deer trachea by comparing extraction rate and extraction time. Finally, dilute alkali-concentrated salt method with high extraction rate and short time was chosen to extract chondroitin sulfate from that. High performance liquid chromatography(HPLC) was used to determined chondroitin sulfate content in deer trachea, and antioxidant activity of chondroitin sulfate and chondroitin sulfate oligosaccharides from deer tracheal was investigated. The results showed that yield of chondroitin sulfate from deer trachea was 21.88% by dilute alkali-concentrated salt extraction, total chondroitin sulfate content in deer trachea was 35.31 g/kg, and chondroitin sulfate A, B, and C contents were 19.54, 2.06 and 13.71 g/kg, respectively. Chondroitin sulfate and chondroitin sulfate oligosaccharides from deer trachea both have certain antioxidant activity, and antioxidant activity of chondroitin sulfate oligosaccharide was significantly higher than that of chondroitin sulfate from deer tracheal.
