西洋参果多糖的纯化及DPPH自由基清除活性研究

摘要
通过水提醇沉法得到西洋参果中总多糖(WQBP),再通过离子交换层析分离纯化,得到中性糖WQBP-N和酸性糖WQBP-A。利用高效液相色谱法对单糖组成进行分析,并检测各级分的1,1-二苯基-2-二硝基苯肼(DPPH)自由基清除活性。结果表明,WQBP主要由鼠李糖(Rhamnose,Rha)、半乳糖醛酸(Galacturonic acid,GalA)、半乳糖(Galactose,Gal)、阿拉伯糖(Arabinose,Ara)和葡萄糖(Glucose,Glc)组成;WQBP-N主要由Gal、Ara、Glc和甘露糖组成,比例为63.9∶19.3∶15.2∶1.5;WQBP-A主要由Rha、GalA、Gal、Ara和Glc构成,比例为13.1∶13.2∶46.4∶19.0∶5.8。WQBP对DPPH自由基清除活性优于其子级分,并且清除率随着浓度的升高而增加。研究结果能够为西洋参果多糖的开发利用提供一定的数据基础。
        Extracted and purified polysaccharide fractions from Panax quinquefolius berry,and DPPH radicals scavenging activity was carried on.Polysaccharide fractions(WQBP) from P.quinquefolius berry was dealed with water-extraction, ethanol-precipetation and ion-exchange chromatography,then one neutral(WQBP-N) and one acidic fraction(WQBP-A) were obtained.The monosaccharide composition,molecular weight distribution and DPPH free radicals scavenging activity were analyzed.WQBP is mainly composed of rhamnose, galacturonic acid, galactose, arabinose and glucose. WQBP-N is composed of galactose, arabinose, glucose and mannose with a ratio of63.9∶19.3∶15.2∶1.5; WQBP-A is mainly composed by rhamnose, galacturonic acid,galactose, arabinose and glucose with a ratio of13.1∶13.2∶46.4∶19.0∶5.8.WQBP has more significant DPPH free radicals scavenging activity than neutral and acidic fraction, which has a concentration dependant tendency.WQBP has potential antioxidant activity in vitro, and the results above could provide theory basis for the further application and developments of the P.quinquefolius berry polysaccharide.
