摘要
以香菇(Lentinula edodes)"沪香F2"为亲本,构建F2代群体,测定85个F2代菌株在PDA平板上的菌丝生长速度、菌包中的菌丝生长速度、原基形成时间、出菇周期、菌盖直径、菌柄直径、菌柄长度、单包产量、单包菇数等农艺性状,利用筛选获得的32对具多态性的SSR引物对F2代群体进行基因型检测,共扩增出93个多态性条带,将观测的9个农艺性状与这93个SSR标记进行相关性分析。结果显示,有26个标记能够与这些性状发生关联,关联事件的总数为30;其中有8个标记表现为极显著关联(P<0.01),2个标记与菌柄长度极显著关联,6个标记与单包菇数极显著关联。本研究首次探索了香菇F2代群体在表型上的分化以及与分子标记之间的关联性,为分子标记辅助育种在香菇中的应用提供参考。
        Lentinula edodes strain ' Huxiang F2 ' was used as the parent to construct a F2 generation population. The mycelial growth rates of 85 F2 strains on PDA and in the bags were determined. Seven other agronomic traits including primordium formation time, fruiting cycle, pileus diameter, stipe diameter, stipe length, fruit body yield per bag and fruit body number per bag were obtained by two times of cultivation.Thirty-two pairs of polymorphic SSR primers obtained after screening were used to dertermine the genotypes of the F2 population and 93 polymorphic bands were obtained. The 9 agronomic traits observed were correlated with these SSR markers. The results showed that there were 26 markers associated with these traits, the total number of associated events was 30, of which 8 markers showed a significant association(P<0.01). Two markers were significantly associated with the stipe length and six markers were significantly associated(P<0.01) with the fruit body number per bag. This study explored phenotypic differentiations of the F2 generation of L. edodes and their association with molecular markers for the first time, which provided a reference for the application of molecular marker-assisted breeding of L. edodes.
