百会、大椎、足三里对癫痫大鼠神经保护机制的影响研究

摘要
目的:研究百会、大椎、足三里对不同机能大鼠海马组织中细胞因子白细胞介素(IL)-1β、IL-2、IL-4、IL-6、IL-10、IL-17、IL-18的表达差异,探讨该针刺方法对海马组织神经保护机制的影响。方法:按随机数字表法将60只雄性SD大鼠平均分为6组:正常空白组、正常经穴组、正常对照组、模型空白组、模型经穴组、模型对照组,每组10只。模型组均采用腹腔注射氯化锂-匹鲁卡品癫痫模型,正常组腹腔注射等量的生理盐水。每组大鼠治疗期间每日固定1次,每次30 min。正常空白组和模型空白组不予针刺只行固定;正常经穴组和模型经穴组,针刺处方为:百会、大椎、足三里;正常对照组和模型对照组,针刺处方为环跳、外关、手三里。治疗10 d后均处死取脑。结果:(1)与正常空白组比较,正常经穴组和正常对照组各细胞因子的指标虽略有差异,但无统计学意义(P>0.05);模型空白组IL-6、IL-18的表达显著上升(P<0.05),IL-1β、IL-2、IL-4、IL-10、IL-17表达无明显差异(P>0.05)。(2)与模型空白组比较,模型经穴组IL-10、IL-18的表达量下降(P<0.05),IL-1β、IL-2、IL-4、IL-6、IL-17的表达差异无统计学意义(P>0.05);模型对照组各组细胞因子的表达无明显差异(P>0.05)。(3)与正常经穴组比较,模型经穴组IL-1β、IL-2、IL-6、IL-18表达量上升(P<0.05), IL-4、IL-10、IL-17的表达无明显差异(P>0.05)。结论:(1)百会、大椎、足三里可能是通过调控癫痫大鼠体内IL-10和IL-18的表达,对癫痫疾病神经机制起到保护作用。(2)百会、大椎、足三里可能是通过影响IL-1β、IL-2、IL-6、IL-18的表达,对不同机能大鼠发挥其双向调节作用。
        Objective:To study the differences of expressions of cytokines IL-1β, IL-2, IL-4, IL-6, IL-10, IL-17 and IL-18 in hippocampus of rats with different functions in Baihui(DU20), Dazhui(DU14) and Zusanli(ST36). To explore the effect of this acupuncture method on the neuroprotective mechanism of hippocampus. Method: Sixty male SD rats were randomly divided into 6 groups according to the random number table method: normal blank group, normal acupoint group, normal control group, model blank group, model acupoint group, model control group, 10 in each group. The model group received an intraperitoneal injection of a lithium chloride-pilocarpine epilepsy model. The normal group received intraperitoneal injection of the same volume of physiological saline. Each group's rats were fixed once a day for 30 minutes. The normal blank group and the model blank group did not need acupuncture and only need to be fixed. The acupuncture prescription of normal acupoint group and model acupoint group: Baihui(DU20), Dazhui(DU14) and Zusanli(ST36). In the normal control group and the model control group, the acupuncture prescriptions were Huantiao(GB30), Waiguan(SJ5), and Shousanli(LI10). After 10 days of treatment, the brains were sacrificed. Result:(1) Compared with the normal blank group, there were slight differences in the cytokines of the normal acupoint group and the normal control group, but there was no statistical significance(P>0.05). The expressions of IL-6 and IL-18 in the model blank group were significant. There was no significant difference in the expressions of IL-1β, IL-2, IL-4, IL-10 and IL-17(P>0.05).(2)Compared with the model blank group, the expressions of IL-10 and IL-18 decreasedin the model acupoint group(P<0.05). There was no significant difference in the expressions of IL-1β, IL-2, IL-4, IL-6 and IL-17(P>0.05). The expression of cytokines in the model control group had no significant difference(P>0.05).(3)Compared with the normal acupoint group, the expressions of IL-1β, IL-2, IL-6 and IL-18 increased in the model acupoint group(P<0.05). There was no significant difference in the expressions of IL-4, IL-10 and IL-17(P>0.05).Conclusion:(1)Baihui(DU20), Dazhui(DU14) and Zusanli(ST36) may protect the neurological mechanisms of epilepsy by regulating the expressions of IL-10 and IL-18 in epileptic rats.(2)Baihui(DU20), Dazhui(DU14) and Zusanli(ST36) may exert their bidirectional effects on different functional rats by affecting the expressions of IL-1β, IL-2, IL-6 and IL-18.
