妊高症患者胎盘MiR-26b和Nkx2-5的表达水平及临床意义

摘要
目的探究妊娠高血压疾病患者胎盘micro RNA-26b(MiR-26b)和Nkx2-5的表达水平及临床意义。方法选择2016年3月~2017年10月于笔者医院接受治疗的60例妊高症患者(包括20例妊娠期高血压、20例轻度子痫前期和20例重度子痫前期)为实验组,另选择同期住院的20例健康孕妇作为对照组。实时定量PCR(q RT-PCR)技术检测胎盘组织MiR-26b水平;蛋白质印迹(Western blot)技术检测胎盘组织Nkx2-5水平。结果轻度子痫前期组和重度子痫前期组的新生儿体重及胎盘重量较健康对照组均显著降低(P<0.05)。q RT-PCR结果显示,妊娠期高血压组、轻度子痫前期组和重度子痫前期组胎盘组织MiR-26b水平较健康对照组均显著升高(P<0.05),且随着病情的加重,MiR-26b水平逐渐上升(P<0.05)。Western blot法检测结果显示,妊娠期高血压组、轻度子痫前期组和重度子痫前期组胎盘组织Nkx2-5水平较健康对照组均显著降低(P<0.05),且随着病情的加重,MiR-26b水平逐渐降低(P<0.05)。妊高症患者胎盘组织MiR-26b水平与Nkx2-5水平显著呈负相关(P<0.05)。妊高症患者胎盘组织MiR-26b水平与新生儿体重、胎盘重量呈负相关(P<0.05),Nkx2-5水平与新生儿体重、胎盘重量显著呈正相关(P<0.05)。结论妊高症患者胎盘组织中MiR-26b表达上调、Nkx2-5表达下调,且与病情严重程度相关,暗示MiR-26b、Nkx2-5可作为妊高症的独立诊断因子。
        Objective To explore the expression level and clinical significance of placenta micro RNA-26 b(MiR-26 b) and Nkx2-5 in patients with pregnancy-induced hypertension.Methods Sixty cases of pregnancy-induced hypertension patients from March2016 to October 2017 treated in author's hospital(including twenty cases of gestational hypertension,twenty cases of mild preeclampsia and twenty cases of severe preeclampsia) were selected as the experimental group,and twenty healthy pregnant women were selected as control group at the same time.Real time quantitative PCR(q RT-PCR) technique was used to detect the placental tissue MiR-26 b level;Western blot technique was used to detect the placental tissue Nkx2-5 level.Results The neonatal weight and placental weigh in mild preeclampsia group and severe preeclampsia group were significantly lower than those in healthy control group(P<0.05).The q RT-PCR results showed that placental tissue MiR-26 b levels in gestational hypertension group,mild preeclampsia group and severe preeclampsia group were significantly higher than the healthy control group(P<0.05),and with the aggravation of the disease,the level gradually increased(P<0.05).The Western blot results showed that gestational hypertension group,mild preeclampsia and severe preeclampsia group placental tissue Nkx2-5 levels were significantly lower than the healthy control group(P<0.05),and with the aggravation of the disease,the level of MiR-26 b decreased gradually(P<0.05).There was a negative correlation between the MiR-26 b level and the Nkx2-5 level in placental tissue of PIH(P<0.05).The placenta tissue MiR-26 b level was negatively correlated with neonatal weight and placental weight(P<0.05),and the Nkx2-5 level was positively correlated with neonatal weight and placental weight(P<0.05).Conclusion The expression of MiR-26 b is up-regulated and the expression of Nkx2-5 is down-regulated in placenta of PIH patients,which are related to the severity of the disease.It suggests that MiR-26 b and Nkx2-5 can be used as independent diagnostic factors for pregnancy-induced hypertension.
